甾体激素类药物与细胞色素P450代谢酶系相互作用的研究进展

【摘要】:甾体激素主要包括肾上腺糖皮质激素、雌激素、孕激素、雄激素和维生素D,临床常用的甾体激素类药物多为其衍生物或人工合成品。细胞色素P450(CYP450)代谢酶系在甾体激素的生物合成及代谢清除过程中起着关键的作用,而甾体激素也可通过对CYP450的诱导或抑制而影响其酶蛋白表达及代谢活性,二者之间的相互作用决定了临床多种疾病(如肿瘤、炎症或自身免疫性疾病)发生的易感性及药物疗效。本文对甾体激素类药物与细胞色素P450代谢酶系相互作用的研究进展进行阐述。
[Abstract]:Steroids mainly include adrenocortical hormone, estrogen, progesterone, androgen and vitamin D. The steroids commonly used in clinic are mostly derivatives or synthetic products of steroids. Cytochrome P450 (CYP450) metabolizing enzymes play a key role in the biosynthesis and metabolic clearance of steroidal hormones. Steroid hormones can also affect the expression and metabolic activity of CYP450 by inducing or inhibiting cytochrome P450 metabolizing enzymes. The interaction between the two determines the susceptibility and drug efficacy of many clinical diseases, such as tumor, inflammation or autoimmune disease. The interaction between steroid hormones and cytochrome P450 metabolic enzymes was reviewed in this paper.
