αB-晶状体蛋白对急性高眼压大鼠视网膜神经节细胞保护作用机制的研究
[Abstract]:Objective to study the effects of 伪 B-crystallin on potassium channel kv1.1,kv1.3, apoptosis inhibitor Bcl-xl and apoptosis factor Caspase-3, and to explore the effect of 伪 B-crystallin on (retinal ganglion cells, in retinal ganglion cells of rats with acute ocular hypertension. RGC) the mechanism of protective effect. Methods Sixty SD rats were randomly divided into group A (), B group with acute hyperocular pressure, group 5 渭 L with intravitreal injection of normal saline). Group C (10 脳 10 g 路L-1 伪 B-crystallin 5 渭 L) received intravitreal injection of 10 脳 10 脳 10 g 路L-1 伪 B-crystallin in group C with 20 eyes in each group, and the right eye was the experimental eye. Western blot and real-time quantitative reverse transcription polymerase chain reaction (QRT-PCR) were used to detect the expression of kv1.1,kv1.3,Bcl-xl and Caspase-3 in retina 7 d and 14 d after injection. Results on the 7th and 14th day after injection, the expression level of kv1.1,kv1.3 channel protein and apoptosis factor Caspase-3 protein and the relative expression of mRNA in group C were lower than those in group A and group B (P 0.05). The expression level of Bcl-xl protein and the relative expression of mRNA in group C were significantly higher than those in group A and group B (P 0.05). At the same time point, there was no significant difference between group A and group B in different factors (P 0.05). Conclusion 伪 B-crystallin can increase the expression of inhibitor of apoptosis factor Bcl-xl and decrease the expression of apoptosis factor Caspase-3 and decrease apoptosis by inhibiting the expression of potassium channel kv1.1,kv1.3 in RGC after acute ocular hypertension. Protect RGC.
