硫酸依替米星含量测定及有关物质反相高效液相色谱-脉冲安培电化学法的建立

【摘要】:目的建立硫酸依替米星原料药含量测定和有关物质检查用的反相高效液相色谱-脉冲安培电化学(LC-PED)方法。方法以15 m L·L-1三氟乙酸溶液(含500μL·L-1五氟丙酸,1.5 g·L-1无水硫酸钠,8 m L·L-150%氢氧化钠溶液,用50%Na OH调节p H值至3.5)-乙腈(96∶4)为流动相,流速1.0 m L·min-1,脉冲安培电化学检测器检测,四波形检测电位,柱温35℃,检测器温度35℃,柱后加0.8 mol·L-1Na OH溶液,流速0.4 m L·min-1。结果 22个杂质得到分离,可将2010年版《中国药典》方法中无法分离的部分杂质有效分离;依替米星在0.24~45μg·m L-1内线性关系良好(r=0.999 7);方法的LOD(S/N=3)为2 ng,LOQ(S/N=10)为6 ng;进样精密度RSD(n=6)为0.8%,含量测定的重复性RSD(n=6)为0.7%,中间精密度RSD(n=18)为0.98%;有关物质的杂质总量重复性的RSD(n=6)为1.72%,杂质总量中间精密度RSD(n=18)为1.71%;溶液在12 h内稳定。结论该方法专属性好,线性范围宽,灵敏度高,重复性优,已经被2015年版《中国药典》收载。
