碳酸二乙酯对大鼠生育力与早期胚胎发育的毒性作用

【摘要】:目的观察碳酸二乙酯(DEC)对雌雄大鼠生育力与早期胚胎发育的影响。方法雌雄SD大鼠分别经口给予DEC 80、200和500 mg/kg·bw·d及阴性对照(大豆油,10 ml/kg·d),雄鼠连续给药4周、雌鼠连续给药2周后按1∶1合笼,交配成功后雄鼠继续给药至剖解,雌鼠继续给药至妊娠第6天(GD6)。试验期间观察各组雌雄SD大鼠的一般毒性反应、体重及摄食量影响,合笼期间雌鼠动情周期,记录交配情况;交配成功雌鼠于GD15剖检,进行终末检查,评价雌性生殖机能、生育力,以及子代的胚胎形成和早期发育影响;雄鼠于交配成功后剖检,并分析精子浓度及活力,评价雄性生殖机能、生育力影响。结果各剂量组DEC对雌雄大鼠亲代均未出现明显的亲代一般毒副反应;对雌雄大鼠体重未见明显异常,仅高剂量GD0和低剂量GD0、GD6雌鼠体重略低于阴性对照组(P0.05);对雌雄大鼠的摄食量、生殖系统脏器重量和脏器系数、交配成功天数、交配率、妊娠率、雌鼠动情周期、雄鼠精子浓度及活力均未见明显影响;对孕鼠子代胚胎形成和早期发育未见明显影响。结论在本试验条件下,雌雄SD大鼠的生育力以及早期胚胎发育均未见明显毒性影响,未观察到明显毒性反应剂量(NOEAL)为500 mg/kg·bw·d。
[Abstract]:Objective to observe the effect of diethyl carbonate (DEC) on fertility and early embryonic development in male and female rats. Methods male and female SD rats were given DEC 80200 and 500 mg/ kg 路bw 路d and negative control (soybean oil, 10 ml/ kg 路d), male rats for 4 weeks). The female rats were given DEC 80200 and 500 mg/ kg 路d), 路d respectively for 4 weeks, and the female rats were caged at 1:1 after 2 weeks of continuous administration. After successful mating, the male rats continued to be given until the dissecting, and the female mice continued to be given until the 6th day of pregnancy (GD6). During the experiment, the general toxic reaction, body weight and food intake of male and female SD rats were observed, the estrous cycle of female rats during cage period was observed, and the mating situation was recorded. The successful mating female mice were examined by GD15, and the female reproductive function, fertility, embryo formation and early development of offspring were evaluated by terminal examination. The male mice were examined after successful mating, and the sperm concentration and motility were analyzed to evaluate the effects of male reproductive function and fertility. Results DEC in each dose group had no obvious toxic and side effects on the parents of male and female rats, but there was no significant abnormality in the body weight of male and female rats, but the body weight of female rats with high dose GD0 and low dose GD0,GD6 was slightly lower than that of negative control group (P 0.05). There were no significant effects on food intake, reproductive system organ weight and organ coefficient, mating success days, mating rate, pregnancy rate, estrus cycle, sperm concentration and motility of male and female rats. There was no significant effect on embryo formation and early development of pregnant rats. Conclusion under the experimental conditions, there is no obvious toxic effect on fertility and early embryonic development in male and female SD rats, and the obvious toxic reaction dose (NOEAL) is 500 mg/ kg 路bw 路d.
