河蚌多糖抗HBV活性及连分式插值法检测

【摘要】:目的探讨河蚌多糖(AP)体外抗乙肝病毒(HBV)的生物活性及量效关系。方法以Hep G2.2.15细胞为体外细胞模型,与1:10梯度稀释的AP 37℃孵育9d,ELISA法检测细胞上清中HBsAg、HBeAg的含量,荧光定量PCR检测HBV DNA载量。采用Thiele型连分式插值法检测AP半数抑制浓度(IC_(50))、最大抑制率并绘制量效曲线图。结果 AP在体外可显著抑制Hep G2.2.15细胞的HBsAg和HBeAg分泌以及HBV DNA复制,且呈现剂量依赖关系。通过Thiele型连分式插值法得出量效关系表达式,并由此确定AP对HBeAg和HBsAg分泌的最大抑制率分别为47.7%和56.4%(抑制HBeAg的IC_(50)为143.7mg/L);对HBV DNA复制的最大抑制率为17.8%。结论 AP具有体外抗HBV生物活性。Thiele型连分式插值法简单、实用,是一种分析药物活性的新方法。
