盐酸帕罗西汀对卵巢切除大鼠学习记忆能力及血清雌激素水平的影响

【摘要】:目的探讨抗抑郁剂盐酸帕罗西汀对卵巢切除大鼠学习记忆能力、血清雌激素水平及大鼠海马雌激素受体表达的影响。方法采用SD雌鼠双侧卵巢切除(OVX)作为雌激素波动引起的抑郁与认知障碍模型,用Morris水迷宫实验来测试大鼠的学习记忆功能。实验共分为正常对照组、OVX对照组、OVX药物组。予5-羟色胺重摄取抑制剂类药物:盐酸帕罗西汀10 mg/(kg·d)干预4周。采用ELISA法测量SD大鼠血清黄体生成素、卵泡刺激素、雌激素。免疫组化法检测大鼠海马雌激素受体α(ERα)和雌激素受体β(ERβ)的阳性细胞表达数。结果 1 Morris水迷宫实验中,OVX对照组和OVX药物组的逃避潜伏期明显缩短,停留在平台时间百分比明显增加。2 OVX药物组的血清雌激素较OVX对照组明显增加(P0.05)。3药物组与非药物组的ERα、ERβ表达无明显差异。结论帕罗西汀增加OVX大鼠血清雌激素水平和改善学习记忆能力,而对海马ERα、ERβ表达无明显影响。
[Abstract]:Objective to investigate the effects of paroxetine hydrochloric acid on learning and memory ability, serum estrogen level and estrogen receptor expression in hippocampus of ovariectomy rats. Methods bilateral ovariectomy (OVX) was used as a model of depression and cognitive impairment induced by estrogen fluctuation in SD female rats. Morris water maze test was used to test the learning and memory function of rats. The experiment was divided into normal control group, OVX control group and OVX drug group. 5-hydroxytryptamine reuptake inhibitor: paroxetine hydrochloric acid 10 mg/ (kg 路d) was given for 4 weeks. Serum luteinizing hormone, follicular stimulating hormone and estrogen were measured by ELISA method in SD rats. The expression of estrogen receptor 伪 (ER 伪) and estrogen receptor 尾 (ER 尾) in rat hippocampus was detected by immunohistochemistry. Results 1 in Morris water maze test, the escape latency of OVX control group and OVX drug group was significantly shortened and the percentage of staying on the platform was significantly increased. 2 the serum estrogen level in OVX drug group was significantly higher than that in OVX control group (P 0.05). 3 there was no significant difference in the expression of ER 伪 and ER 尾 between drug group and non-drug group. Conclusion Paroxetine increases serum estrogen level and improves learning and memory ability in OVX rats, but has no significant effect on the expression of ER 伪 and ER 尾 in hippocampus.
