Pestalachloride E,红树林内生杂色拟盘多毛孢真菌中的新氯代二苯甲酮衍生物(英文)

【摘要】:内生真菌已经成为发现结构新颖、活性显著的次级代谢产物的重要来源,红树林内生真菌的代谢产物中发现了许多结构多样的化合物,引起了人们的广泛重视。采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、以及制备薄层色谱等方法从南海红树林植物桐花树内生杂色拟盘多毛孢真菌大米发酵产物中的乙酸乙酯提取物中得到4个代谢产物,其中1个新氯代二苯甲酮衍生物pestalachloride E(2),2个已知的二苯甲酮衍生物pestalachlorides C(1)和B(3),1个已知的蒽醌化合物,大黄素甲醚physcion(4)。新化合物通过一维和二维核磁共振谱鉴定,并通过高分辨电喷雾质谱确认。细胞毒实验结果表明化合物1显示良好的抗肿瘤活性,IC50在2.8~82.4μmol/L区间。化合物3显示具有抗炎活性,对环氧合酶-2的抑制活性为IC50 2.4μmol/L。
[Abstract]:Endophytic fungi have become an important source of the secondary metabolites with novel structure and active activity, and many compounds with various structures are found in the metabolites of the mangrove endophytic fungi. adopting silica gel column chromatography, Sephadex LH-20 gel column chromatography, and preparing a thin-layer chromatography to obtain 4 metabolites from the ethyl acetate extract in the rice fermentation product of the endogenous variegated polychaete chrysosporium in the mangrove plant of the South China Sea, One of the new chlorobenzophenone derivatives, pestalachloride E (2), two known benzophenone derivatives, pestalachlorides C (1) and B (3),1 known dibenzo compound, emodin methyl ether phycion (4). The new compound was identified by one-dimensional and two-dimensional nuclear magnetic resonance spectroscopy and confirmed by high-resolution electrospray mass spectrometry. The results of the cytotoxicity test show that the compound 1 shows good anti-tumor activity and the IC50 is in the range of 2.8-82.4. m Compound 3 showed an anti-inflammatory activity and an inhibitory activity on cyclooxygenase-2 was IC50 of 2.4. m u.mol/ L.
