体外和体内磷酸肌酸可降低血糖水平,改善肝细胞内质网应激的糖尿病小鼠

背景肝细胞受损可增加糖尿病(Diabetes mellituspatients,DM)患者发病率和死亡率。已公认,高血糖是DM患者肝损害发病的主要原因,与DM相关的肝细胞损害是DM患者常见的并发症。磷酸肌酸(Phosphocreatine,PCr)分子作为一种可迅速动员、回收ATP,对脑和骨骼肌的能量储备,可作为细胞能量储备形式,它具有多种活性。然而,PCr是否具有保护DM相关肝损伤的作用尚未见报道。本研究旨在评价PCr是否能保护DM并发的肝损伤,并探讨其在体内外作用机制。材料与方法体外研究:丙酮醛(methylglyoxal,MGO),是葡萄糖的活性代谢产物,与高血糖可引起DM相关的肝细胞损伤,且与肝细胞损害进展关系密切。Hep G2细胞培养及细胞进行预处理MGO两小时,然后,分别用不同浓度的PCR处理细胞(5m M,10m M,20m M)。采用MTT法研究胞形态学、细胞毒性、细胞凋亡;流式细胞仪Annexin V-FITC、Calcien法分别检测细胞凋亡、增殖试验。通过Western blotting检测凋亡蛋白Bax、Bcl-2和Caspase-3、Caspase-9蛋白表达...
