一种萘啶衍生物3x通过非Caspase凋亡途径诱导A375细胞死亡(英文)

萘啶化合物是一类具有多种生理学特性的药物结构单元,在医学、药学、生理学等诸多领域都有着广泛的应用.一种新的化合物(10-甲氧基-1,2,3,4-四氢苯并[g][1,3]二氮杂卓[1,2-a][1,8]萘并吡啶-6-基)(4-甲基环己-2,4-二烯-1-基)甲酮(命名为3x),经噻唑蓝四唑溴化铵(MTT)检测,在人恶性黑色素瘤细胞株A375中显示出良好的抗癌活性和选择性.通过免疫印迹法的验证,我们发现尽管3x可以明显降低XIAP的表达量,但其并不通过激活Caspase-3,-8,-9等蛋白来诱导A375细胞死亡,而是极有可能通过引起p53的增加以及p53的磷酸化来致使Bax、PUMA、AIF的表达量增加,最终导致A375细胞死亡.
