猪皮胶原蛋白酶解物的抗氧化特性研究

【摘要】:本论文以猪皮为原料提取胶原蛋白并制备出胶原蛋白酶解物,分别对其物理性质、不同分子量肽段组分的体外抗氧化活性、体内抗氧化作用以及作为抗氧化剂在猪油及猪肉糜中的抗氧化效果进行研究,为进一步利用猪皮胶原蛋白开发抗氧化肽提供试验基础。 1.分析猪皮胶原蛋白酶解物的溶解性、吸油性、乳化性、乳化稳定性、起泡性及起泡稳定性等物理性质,为其在食品、保健品等领域的有效利用提供一定的依据。 2.利用不同膜截留分子量的超滤离心管对猪皮胶原蛋白酶解物进行分离,得到分子量大于10kDa、5kDa-10kDa、3kDa-5kDa和小于3kDa的4个分子量肽段的组分,以氧自由基(02-·)清除活性、羟自由基(·OH)清除活性、DPPH·自由基清除活性、还原力、脂质过氧化抑制能力为指标,测定不同分子量肽段组分及酶解液的抗氧化能力。结果显示,不同分子量范围的猪皮胶原蛋白酶解物均具有一定的抗氧化能力,且随着浓度的增大,其抗氧化活性越强。其中,酶解液对O:-·清除活性最好,分子量5kDa-10kDa组分对·OH清除活性、还原力和脂质过氧化抑制能力最好,3kDa-5kDa组分对DPPH·自由基清除活性最好。 3.将昆明小鼠随机分成5组,分别为:空白组、衰老模型组、猪皮胶原蛋白酶解物低剂量组(100mg/kg).猪皮胶原蛋白酶解物中剂量组(200mg/kg)和猪皮胶原蛋白酶解物高剂量组(400mg/kg)。除空白组外,其他各组均颈背部皮下注射D-半乳糖,饲养42d后,以小鼠血清、肝脏和大脑中的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)的活性及丙二醛(MDA)含量为指标,考察猪皮胶原蛋白酶解物在小鼠体内的抗氧化性能。结果表明,与空白组小鼠相比,衰老模型组小鼠的血清、肝脏、大脑中的SOD、GSH-PX活性明显降低,MDA含量明显升高;猪皮胶原蛋白酶解物能提高小鼠血清、肝脏和大脑中的SOD和GSH-PX活性,降低MDA含量,并呈现出一定的剂量效应关系。猪皮胶原蛋白酶解物对D-半乳糖致衰老小鼠具有一定的抗氧化作用。 4.将不同量的猪皮胶原蛋白酶解物分别添加到猪油及猪肉糜中,考察其对猪油及猪肉糜氧化的抑制作用。结果表明,猪皮胶原蛋白酶解物对猪油及猪肉糜均具有一定的抗氧化作用,且随着添加量的增加其抗氧化能力也增强,当添加量为2%时,其抗氧化效果最好,但其效果略低于0.02%的叔丁基羟基茴香醚(BHA)。
[Abstract]:In this paper, collagen was extracted from pig skin and collagen proteolysis was prepared. The physical properties and antioxidant activity of different molecular weight peptide fractions in vitro were studied. The antioxidative effect in vivo and antioxidant activity in pork and lard were studied, which provided the experimental basis for the further development of antioxidant peptides by using porcine skin collagen. 1. The solubility, oil absorption, emulsification, emulsification stability, foaming property and foaming stability of porcine skin collagen protease hydrolysate were analyzed, which provided some basis for its effective utilization in food and health products. 2. The porcine skin collagen proteolytic compounds were separated by ultrafiltration centrifuge tubes with different molecular weight, and the four molecular weight peptides with molecular weight larger than 10kDa5kDa-10kDa-5kDa and less than 3kDa were obtained. Hydroxyl radical scavenging activity DPPH 路free radical scavenging activity, reducing capacity and inhibiting capacity of lipid peroxidation were used as indexes to determine the components of different molecular weight peptides and the antioxidant ability of enzymatic hydrolysis solution. The antioxidation activity of porcine skin collagen protease hydrolysates with different molecular weight ranges was higher with the increase of concentration, among them, the enzymatic hydrolysis solution had the best scavenging activity against O:-路, and the molecular weight of 5kDa-10kDa component had the best scavenging activity against 路OH. Reduction and lipid peroxidation inhibition were the best. The 3kDa-5kDa component had the best scavenging activity against DPPH 路free radical. 3. Kunming mice were randomly divided into five groups: blank group, aging model group, low dose group of porcine skin collagen proteolytic substance (100mg / kg), middle dose group (200mg / kg) and high dose group (400mg / kg). Dgalactose was injected subcutaneously into the neck and back of other groups. After 42 days of feeding, the activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and the content of malondialdehyde (MDA) in serum, liver and brain of mice were used as indexes. The antioxidative activity of porcine collagen protease hydrolysate in mice was investigated. The results showed that the activity of SOD- GSH-PX in serum, liver and brain of aging model group was significantly lower than that of blank group. Porcine skin collagen proteolytic agent can increase the activity of SOD and GSH-PX in serum, liver and brain of mice, and decrease the content of MDA. The porcine skin collagen proteolysis had a certain antioxidant effect on aging mice induced by D-galactose. 4. The different amounts of porcine skin collagen protease hydrolysates were added to the pork and lard respectively, and the inhibitory effect on the oxidation of the lard and the pork surimi was investigated. Porcine skin collagen protease hydrolysate had antioxidation effect on both lard and pork chilli, and its antioxidant activity also increased with the increase of the amount of pig skin collagen protease, and the antioxidant effect was the best when the amount of collagen protease hydrolysate was 2%. But its effect is slightly lower than that of uncle Ding Ji hydroxyl anisole (0.02%).
