野木瓜对镉致SD大鼠亚慢性肝、肾损伤保护作用机制初探

【摘要】:目的:探讨野木瓜(Stauntonia chinensi,SC)对镉致SD大鼠亚慢性肝肾损伤的保护作用及其可能机制。方法:雄性SD大鼠50只,进行体重分层,根据随机数字法分为5组,每组10只。分组如下:空白对照组(C组)、单独染镉组(Cd组)两组每天灌胃与体重相应的生理盐水,野木瓜低、中、高剂量组(SCL:0.5 g/kg;SCM:1g/kg;SCH:2g/kg)每天灌胃相应剂量野木瓜悬浊液,除C组外,其余4组在饮水中添加Cd Cl2(50mg/L),饲养90天后处死大鼠。饲养结束后麻醉取血、肝、肾,-80℃保存。血生化检测肝、肾功能相关指标;测定肝、肾MDA、SOD、GSH-Px含量;原子吸收光谱法检测肝、肾组织及对应组织超微结构镉含量;观察肝、肾脏病理以及超微结构改变;实时荧光PCR检测肝、肾Mt-1、Mt-2 m RNA表达情况;Western blot检测MT蛋白相对表达量。结果:染镉之后,Cd组肝、肾脏器系数均高于C组(P0.05);SCH组肝、肾脏器系数均较Cd组低(P0.05),SCH组效果优于SCL组(P0.05);与C组相比,Cd组ALT、AST、肌酐、尿酸大幅升高(P0.05),SCM、SCH两组ALT、AST、肌酐、尿酸均有不同程度降低(P0.05);肝脏病理结果显示:与C组相比,Cd组肝脏细胞发生肿胀,细胞核固缩,炎性细胞浸润和坏死,野木瓜干预能明显减轻镉诱发的炎症反应,减少肝细胞坏死,改善肝脏损伤;肾脏病理结果发现:与C组相比,Cd组肾小球萎缩,肾小管上皮细胞发生肿胀,肾小管管腔变小,肾小管上皮细胞出现缺失坏死,野木瓜干预明显减轻肾损伤,改善肾小管上皮细胞肿胀。染镉之后,Cd组肝超微结构的损害主要集中在线粒体以及内质网上,表现为内质网水肿,线粒体嵴内隙水肿扩张;Cd组肾超微结构显示,肾小球局部可见足细胞次级突起分布不均匀,基膜厚度不均匀;野木瓜各干预组能观察到镉对肝、肾超微结构的损害有所减轻;原子吸收结果显示:与Cd组相比,SCM、SCH两组中肝、肾、线粒体、微粒体镉含量明显减少(P0.05);相较于Cd组,野木瓜干预组线粒体、微粒体中镉含量减少,并且野木瓜干预组细胞质镉含量高于单独染镉组(P0.05);Cd组肝脏、肾脏脂质过氧化指标MDA含量升高,SOD、GSH-Px活性明显降低(P0.05),给予野木瓜干预后,SCM、SCH两组MDA含量均降低(P0.05),SOD、GSH-Px均有不同程度升高(P0.05)。RT-PCR检测相关基因发现:与Cd组相比,野木瓜干预各组肝、肾Mt-1 m RNA、Mt-2 m RNA表达增高(P0.05),且SCM、SCH组增高更为明显;肝、肾组织MT蛋白印记结果显示:与Cd组相比,SCM、SCH组MT蛋白相对表达显著升高(P0.05)。结论:野木瓜对镉致SD大鼠肝肾损伤具有保护作用。
[Abstract]:Objective: to investigate the protective effect and possible mechanism of Stauntonia chinensis SC) on subchronic liver and kidney injury induced by cadmium in SD rats. Methods: 50 male SD rats were divided into 5 groups according to random number method.
