超氧化物歧化酶传递体的制备及体外透皮实验

【摘要】:目的: .探索制备超氧化物歧化酶传递体的合适工艺,分析其理化特征,最后考察其透皮吸收情况。 方法: 以薄膜-挤压法制备超氧化物岐化酶(SOD)传递体,以BCA蛋白浓度检测法测定其包封率。采用微量连苯三酚自氧化法(325nm法)来评价超氧化物歧化酶的活性。另外,考察了SOD传递体在大鼠皮肤的体外经皮吸收情况。结果: 制备了粒径均一的SOD传递体制剂。用BCA蛋白检测法测定SOD浓度,在5-1000μg/ml的浓度区间内,具有较好的线性。制备的传递体包封率为(57.3±0.5)%,平均粒径是(158.4±12.5)nm,与SOD溶液以及普通脂质体相比,SOD传递体能透过皮肤,而前两者未检测到SOD。 结论: 该超氧化物歧化酶传递体制备方法可行,其分析方法稳定。制备的超氧化物歧化酶传递体有较高的包封率,,并能透过大鼠的皮肤。
[Abstract]:Objective:
A suitable process for preparing superoxide dismutase (SOD) transfer body was explored, and its physicochemical characteristics were analyzed. Finally, the transdermal absorption of the superoxide dismutase was investigated.
Method:
The film prepared by extrusion of superoxide dismutase (SOD) transfer, the encapsulation rate was determined by BCA assay. The protein concentration of Trace Benzene even three phenolic autoxidation method (325nm method) to evaluate the superoxide dismutase activity. In addition, the effects of SOD transfer in rat skin in vitro percutaneous absorption results:
The preparation of uniform size SOD transfer agent system. The concentration of SOD protein was detected with BCA, in the concentration range of 5-1000 g/ml, with good linearity. The preparation of transfersome encapsulation was (57.3 + 0.5)%, the average particle size is (158.4 + 12.5) nm, compared with SOD solution and ordinary liposomes, SOD physical transmission through the skin, and both were not detected by SOD.
Conclusion:
The preparation method of the superoxide dismutase delivery system is feasible, and its analytical method is stable. The prepared superoxide dismutase transporter has high encapsulation efficiency and can penetrate rat skin.
