DEHP对CHO细胞凋亡基因和癌基因表达水平的影响

【摘要】:目的探讨DEHP对中国仓鼠卵巢细胞(CHO细胞)的细胞毒性和凋亡基因癌基因表达水平的影响。方法不同剂量DEHP对CHO细胞进行染毒24 h和48 h,应用CCK8试剂盒测定DEHP对CHO细胞的半数抑制浓度IC50,荧光定量PCR检测凋亡基因(Bcl-2、caspase-8、caspase-9)和癌基因(c-fos、k-ras、p53)表达水平改变。结果DEHP染毒CHO细胞24 h的IC50为612μM,染毒48 h的IC50为336μM。DEHP较高剂量(150μM和300μM)染毒条件下,凋亡基因Bcl-2表达水平比对照组显著降低,300μM DEHP染毒时caspase-8和caspase-9表达水平比对照组显著增加;150μM和300μM DEHP染毒条件下c-fos表达水平增加;300μM DEHP染毒条件下k-ras基因表达水平升高;p53表达水平无明显变化。结论 DEHP可通过激活caspase通路引起CHO细胞凋亡,也引起癌基因表达水平升高。
[Abstract]:Objective to investigate the effects of DEHP on cytotoxicity and expression of apoptotic gene oncogene in Chinese hamster ovarian cell (Cho). Methods CHO cells were exposed to different doses of DEHP for 24 h and 48 h. The IC50 concentration of DEHP on CHO cells was measured by CCK8 kit. The expression of apoptotic gene (Bcl-2caspase-8 caspase-9) and oncogene c-fosk-rasp53 were detected by fluorescence quantitative PCR. Results the IC50 of CHO cells exposed to DEHP for 24 h was 612 渭 M, and the IC50 of 48 h was 336 渭 M.DEHP. The expression of apoptosis gene Bcl-2 was significantly lower than that of control group. The expression level of caspase-8 and caspase-9 was significantly higher than that of control group under 300 渭 M DEHP exposure. The expression level of c-fos was significantly increased under the condition of 150 渭 M and 300 渭 M DEHP exposure. The expression level of k-ras gene was increased under the condition of 300 渭 M DEHP exposure. The expression level of k-ras gene was increased. There was no significant change in expression level. Conclusion DEHP can induce apoptosis of CHO cells and increase the expression of oncogene by activating caspase pathway.
