DEHP、MEHP对小鼠胚胎干细胞细胞毒性的研究

【摘要】:目的分析不同浓度的邻苯二甲酸二乙基己酯[di(2-ethylhexyl)phthalate,DEHP]及其活性代谢产物邻苯二甲酸单乙基己基酯[mono(2-ethylhexyl)phthalate,MEHP)对小鼠胚胎干细胞细胞毒性、增殖的影响。方法用含有不同浓度DEHP(1 000、100、10、1、0.1、0.01μmol/L)、MEHP(1 000、100、10、1、0.1、0.01μmol/L)的培养液,对胚胎干细胞进行培养,用CCK-8法检测细胞活性。结果染毒96 h后,高剂量(1 000μmol/L)DEHP、MEHP均对胚胎干细胞的增殖有抑制作用(P0.05),且染毒剂量与细胞抑制率之间存在明显的正相关性,相关系数分别为0.51、0.62(P0.05)。结论在一定浓度范围内,DEHP及MEHP均对小鼠ESC具有细胞毒性,且有浓度依赖性。
[Abstract]:Objective to study the effects of di (2-ethylhexyl) phthalate,DEHP and its active metabolite, mono (2-ethylhexyl) phthalate,MEHP, on the toxicity and proliferation of mouse embryonic stem cells. Methods embryonic stem cells were cultured in culture medium containing different concentrations of DEHP (1 000 DEHP (0. 1 0. 01 渭 mol/L), MEHP (1 000) and 0. 1 0. 01 渭 mol/L). The activity of embryonic stem cells was detected by CCK-8 method. Results after 96 h exposure, high dose (1 000 渭 mol/L) DEHP,MEHP could inhibit the proliferation of embryonic stem cells (P0.05), and there was a significant positive correlation between the dose and the cell inhibition rate, the correlation coefficient was 0.51 渭 mol/L (P0.05). Conclusion in a certain concentration range, both DEHP and MEHP have cytotoxicity to ESC in a concentration dependent manner.
