雪莲果汁对高脂模型大鼠的降脂作用及其机制初步探讨

【摘要】:观察雪莲果对高脂高糖致高脂大鼠模型的降脂作用,并对其影响机制进行初步探讨。取SD雄性大鼠喂养高脂高糖饲料建立高脂模型成功后,将高脂模型大鼠随机分为阴性对照组,阳性对照组,雪莲果高、中、低剂量组。各组动物按各药物和剂量灌胃给药至第16周后测定血清血脂水平及肝脏AMPK和ACC在基因和蛋白表达水平。与阴性对照相比,高、中剂量组TG、TC及LDL-C含量降低,HDL-C含量升高;AMPK的基因和蛋白表达水平高于阴性对照组;ACC的基因和蛋白表达水平低于阴性组;低剂量组与阴性对照比无明显差异;一定剂量的雪莲果汁能上调大鼠肝脏组织中AMPK m RNA和蛋白质表达,下调ACC m RNA和蛋白质表达,从而抑制脂肪酸合成,同时加速脂肪酸氧化,达到降脂作用。
[Abstract]:To observe the hypolipidemic effect of Saussurea nucifera fruit on hyperlipidemic rat model induced by high fat and high sugar, and to explore its mechanism. After SD male rats were fed with high fat and high sugar diet, the rats were randomly divided into negative control group, positive control group, high, middle and low dosage group of Saussurea nucifera fruit. The serum lipids and the expression of AMPK and ACC in the liver were measured by intragastric administration of various drugs and doses until the 16th week. Compared with the negative control group, the content of TG,TC and LDL-C decreased and the content of HDL-C increased in the middle dose group, the gene and protein expression of AMPK was higher than that of the negative control group, the expression level of ACC gene and protein was lower than that of the negative group. There was no significant difference between low dose group and negative control group. A certain dose of Saussurea nucifera juice could up-regulate the expression of AMPK m RNA and protein, down-regulate the expression of ACC m RNA and protein, thus inhibit the synthesis of fatty acids, and accelerate the oxidation of fatty acids to achieve the lipid-lowering effect.
