马尾松花粉提取物对前列腺肥大小鼠的影响

【摘要】:前列腺肥大(BPH)是老年男性的常见疾病,为患者的身体和心灵造成了很大的损害。目前治疗BPH的主流药物为5α-还原酶抑制剂和α1-肾上腺素受体阻滞剂,但它们均存在一定的副作用。马尾松花粉提取物(MPPE)是一种具有良好的抗炎和抗氧化活性的天然植物提取物,其营养丰富,无毒副作用。很多研究表明,MPPE可用于改善BPH。本实验拟采用腹腔注射丙酸睾酮(5mg/kg·d)的方法建立小鼠BPH模型,通过探究MPPE以及SQ、PGY两种复方对小鼠BPH的影响,为松花粉进一步开发为抗BPH药物或保健品提供理论依据。实验包括以下几部分: 第一,,MPPE对小鼠BPH的预防作用探究:实验周期为6周,前两周三个松花粉组小鼠预先灌胃相应剂量的MPPE。后四周,在腹腔注射丙酸睾酮建立BPH模型的同时给予三种不同剂量(20、40、80mg/kg)的MPPE。实验结束时收集小鼠尿液,进行代谢组学分析,然后处死小鼠收集血清测定相关生理指标,剖取前列腺制作石蜡切片,观察组织形态变化。 结果显示,与空白组相比,模型组小鼠的前列腺重量、前列腺指数显著增加,前列腺上皮细胞增生,向腺腔内折叠呈绒毛状,形态结构具有明显的BPH特征。三种剂量的MPPE作用效果有一定差异。与模型组相比,20mg/kg MPPE保护作用较小,而40、80mg/kgMPPE组的前列腺指数、炎性细胞因子含量、DHT浓度及PACP、NOS的活性均有明显的降低,NO的生成也相应减少。其中,80mg/kg的MPPE效果最好。PLS-DA结果与生化指标基本一致,对照组与模型组小鼠完全分开,说明BPH小鼠的尿液代谢物发生了明显的变化。非那雄胺组最接近空白组,MPPE的保护作用比较明显,但并未使小鼠完全恢复健康。 第二,MPPE对小鼠BPH的治疗作用探究:与预防作用部分实验不同的是,治疗作用实验先腹腔注射丙酸睾酮4周建立BPH模型。然后再分别灌胃不同浓度的MPPE治疗4周。结果显示,模型组小鼠的前列腺重量、前列腺指数显著增加,前列腺组织形态具有明显的BPH特征。20mg/kg MPPE作用较小,80mg/kg的MPPE效果最好。BPH小鼠的尿液代谢物发生了明显的变化,MPPE作用效果比较明显,但并未使小鼠完全恢复健康。非那雄胺对BPH的改善效果最佳。 另外,对PI百分抑制率进行对比,预防试验中三个剂量MPPE的抑制率分别为26.09%、52.17%、69.57%,而治疗实验的则分别为33.33%、57.14%、76.19%,可以看出MPPE对BPH的预防作用优于其治疗作用。 第三,MPPE及SQ、PGY复方对小鼠BPH的影响:实验具体步骤与MPPE治疗作用实验一致,1-4周:建模;5-8周:治疗。实验结束后测定相关指标。实验结果显示,SQ、PGY两个复方的抗BPH活性明显优于MPPE。且SQ的效果比PGY稍好,但两者差异没有统计学意义。 以上结果表明,(1)MPPE对BPH的预防作用优于其治疗作用,推测可能的原因是因为一边建模一边给药,MPPE及时的发挥了保护作用,减少了小鼠患BPH的可能;(2)SQ、PGY复方的效果优于MPPE,且SQ的效果比PGY稍好。说明SQ、PGY两种中草药与MPPE复合后可以起到一定的强化作用,能更好的改善BPH病症,对机体产生显著的保护作用;(3)MPPE可以降低BPH病理症状及相关生化指标,调节标志性代谢物的水平,且具有一定的剂量依赖性,MPPE的抗BPH作用机理可能与其抗炎抗氧化活性有关。
[Abstract]:Prostatic hypertrophy (BPH) is a common disease in the elderly, causing great damage to the body and mind of the patient. Currently, the main drug for treating BPH is a 5-dose reductase inhibitor and a 1-epinephrine receptor blocker, but all of them have certain side effects. Pinus massoniana pollen extract (MPPE) is a natural plant extract with good anti-inflammatory and antioxidant activity, and has rich nutrition and no toxic and side effects. Many studies have shown that MPPE can be used to improve BPH. In this experiment, a mouse BPH model was established by means of intraperitoneal injection of testosterone propionate (5 mg/ kg 路 d), and the effect of two compounds of MPPE and SQ and PGY on the BPH of mice was investigated.
