海洋芽孢杆菌中Iturin A分离鉴定研究性实验教学探索

【摘要】:以一株海洋Bacillus sp.细菌为对象,进行天然药物化学研究性实验的教学探索。通过优化菌株发酵条件、改进文献提取分离纯化方案,高效率地分离纯化出目标产物Iturin A,并指导学生综合运用质谱,1HNMR结合文献调研鉴定产物结构。该实验合理使用制备高效液相色谱仪,液质联用仪、核磁共振仪等大型仪器设备,改进了传统天然药物化学实验内容,实现了学生实验中微量天然产物快速、高效、可视化的分离与纯化。实验有助于激发学生学习兴趣,使其全面深刻地认识微生物天然产物的科学研究流程,了解掌握不同大型仪器的应用范围和操作技术,提升操作技能和创新能力。
[Abstract]:With a marine Bacillus sp. Bacteria as the object, carry on the natural medicine chemistry research experiment teaching exploration. By optimizing the fermentation conditions of the strain, improving the extraction and purification scheme of the literature, the target product Iturin A was isolated and purified efficiently, and the students were instructed to use mass spectrometry and 1HNMR combined with literature investigation to identify the structure of the product. In this experiment, the preparation of high performance liquid chromatograph, liquid mass spectrometer, nuclear magnetic resonance instrument and other large instruments and equipment were reasonably used, which improved the content of traditional natural drug chemistry experiment and realized the rapid and high efficiency of trace natural products in students' experiment. Visual separation and purification. The experiment is helpful to stimulate students' interest in learning, to have a comprehensive and profound understanding of the scientific research process of microbial natural products, to master the application scope and operation technology of different large-scale instruments, and to improve their operation skills and innovative ability.
