Development and Validation of KASP Markers for Wheat Breedin

功能标记(Functional markers,FMs)在作物育种过程中具有正要价值,功能标记的高通量基因型检测为分子标记辅助选择(Marker-assisted selection,MAS)在育种过程中的应用提供了很好的机会。在本研究中,我们开发并鉴定了一批与普通小麦适应性、产量、品质、生物和非生物胁迫抗性相关的KASP(Kompetitive Allele Specific PCR)标记,并对152个来自于本研究和公共数据库的KASP标记在实际应用中进行可靠性鉴定。我们在以下三个方面对152个KASP标记中的70个进行可靠性验证:1)在23个不同的小麦株系中对开发的KASP标记和基于凝胶的传统PCR标记进行比较,2)利用来自中国和其他13个国家的300份品种的表型数据来验证开发标记等位基因信息的有效性,3)在四个小麦分离群体中对开发的KASP标记进行鉴定。研究结果表明所有的KASP标记都与栽培品种和双亲群体中的相关表型有显著的关联,从而揭示了 KASP标记在小麦育种中潜在的应用前景。本研究表明KASP标记的检测效率是基于凝胶的传统PCR标记的45倍。目前,KASP标记已发展为一种单...
