采用高压均质法制备阿苯达唑纳米脂质体的工艺优化

【摘要】:采用单因素和Box-Behnken设计-效应面法优化了微射流高压均质法制备阿苯达唑纳米脂质体的处方和工艺参数。分别以均质压力、均质温度、阿苯达唑与大豆磷脂质量比为考察对象,以基于平均粒径和包封率的综合指标为评价指标,在单因素考察结果的基础上采用3因素3水平星点设计继续试验,对结果进行二项式拟合,再用效应面法筛选出最佳工艺条件。结果确定最优工艺为:制备温度37℃,均质压力137.90 MPa,均质次数8次,阿苯达唑与大豆磷脂质量比为1∶2。所得纳米脂质体的粒径为(208.25±2.13)nm,包封率为(96.16±0.74)%,粒径与包封率的实测值与预测值的偏差分别为0.94%和0.05%。透射电镜显示纳米脂质体粒径均一,呈球状分布;在室温及4℃放置6个月稳定性良好。
[Abstract]:The formulation and technological parameters of albendazole nanoliposomes were optimized by single factor and Box-Behnken design-effect surface method.The homogenization pressure, homogenization temperature, mass ratio of albendazole to soybean phospholipid were investigated, and the comprehensive indexes based on average particle size and entrapment efficiency were used as the evaluation index.On the basis of the results of single factor investigation, three factors and three levels of star point design were used to continue the experiment, and the binomial fitting was carried out, and the optimum technological conditions were selected by using the effect surface method.The results showed that the optimum conditions were as follows: preparation temperature 37 鈩,
