枸杞多糖对脑力疲劳型心理亚健康模型大鼠认知功能的影响

【摘要】:目的:建立脑力疲劳型心理亚健康大鼠模型,并观察枸杞多糖对脑力疲劳型心理亚健康大鼠认知功能的影响,为心理亚健康的预防与干预提供理论指导。方法:取清洁级Sprague Dawley大鼠50只,按照体重分层随机分为5组,即正常空白对照组(C,n=10),模型空白对照组(PS,n=10),模型给生理盐水组(PS+NS,n=10),模型给枸杞多糖低剂量组(PS+LBP20mg/kg,n=10),模型给枸杞多糖高剂量组(PS+LBP40mg/kg,n=10)。造模程序分为环境适应期、空瓶不确定应激饮水训练期和正式造模三个阶段。采用复合应激因素进行模型的构建,模拟现代社会脑力工作者长期睡眠不足、久坐不动活动量较少、饮食不规律和工作当中不确定出现的应激压力以及人际交往问题来进行模型的构建。从正式造模的第1天起至造模结束的第21天,采用可变计量法分别给予PS+NS组、PS+LBP20mg/kg组和PS+LBP40mg/kg组大鼠相应剂量的灌胃给药。造模结束后,通过鉴别大鼠是否处于疾病状态、进行活动性与情绪的检验和学习记忆实验来检验脑力疲劳型心理亚健康的主要核心症状,对新模型和药物的干预进行检验和评价。结果:1.脑力疲劳型心理亚健康大鼠模型的鉴定C组大鼠与PS组大鼠第7天、第14天、第21天的体重以及体重增长量差异均具有统计学意义(P0.01)。PS组大鼠体重低于C组大鼠,且体重相比C组明显增长缓慢。C组与PS组两组大鼠的旷场垂直站立次数、旷场总距离和平均速度差异显著(P0.05)。两组大鼠的旷场水平运动次数差异显著(P0.01)。C组与PS组两组大鼠的开放臂滞留时间比例方面的差异有统计学意义(P0.05)。C组与PS组两组大鼠在Day6、Day7八臂迷宫的测试完成时间差异具有统计学意义(P0.05)。C组与PS组两组大鼠在Day6八臂迷宫的工作记忆错误次数具有统计学意义(P0.05)。2.枸杞多糖干预对五组大鼠的影响(1)C组21天的体重与PS+LBP40mg/kg组、PS+LBP20mg/kg组PS+NS组和PS组组间比较均具有统计学意义(P0.01)。PS+LBP40mg/kg组、PS+LBP20mg/kg组21天的体重与PS+NS组和PS组组间比较均具有统计学意义(P0.01)。第1周、第2周、第3周的体质量增长量各组大鼠的组间差异均具有统计学意义(P0.01)。(2)C组与PS组的旷场总距离、平均速度、垂直站立次数、水平跨格数比较差异均有统计学意义(P0.05)。PS+LBP20mg/kg组与PS组的旷场总距离、垂直站立次数、旷场水平跨格数比较差异有统计学意义(P0.05)。PS+LBP40mg/kg组与PS组的旷场垂直站立次数、水平跨格数比较差异有统计学意义(P0.05),(3)C组与PS+NS组开放臂进入次数比例、开放臂滞留时间比例均具有统计学意义(P0.05),PS组与C组、PS+LBP40mg/kg组开放臂进入次数比例差异有统计学意义(P0.05)。PS组与C组、PS+LBP40mg/kg组、PS+LBP20mg/kg组开放臂滞留时间比例差异有统计学意义(P0.05)。(4)八臂迷宫测试第3天PS+LBP40mg/kg与PS+NS组的测试时间差异有统计学意义(P0.05)。八臂迷宫测试第6天C组与PS+NS组、PS组参考记忆错误次数比较(P0.05);PS+LBP40mg/kg与PS参考记忆错误次数比较(P0.05);PS+LBP20mg/kg组与PS+NS参考记忆错误次数比较(P0.05)。八臂迷宫测试第7天C组与PS+NS组、PS组参考记忆错误次数比较(P0.05)。八臂迷宫测试第6天C组与PS+NS组、PS组正确率比较(P0.05),PS+LBP40mg/kg与PS组正确率比较(P0.05)。结论:1.通过慢性不完全睡眠剥夺、活动限制、饮食不规律、空瓶不确定性心理应激和社交失败复合应激因素进行21天的脑力疲劳型心理亚健康大鼠模型的构建,从一般情况观察、体温、体重、旷场实验、高架十字迷宫实验和八臂迷宫实验检测指标的分析结果认为,脑力疲劳型心理亚健康模型的构建基本成功。2.脑力疲劳型心理亚健康模型大鼠的症状表现主要为体重增长缓慢、精神紧张或烦躁不安造成的自主活动性活动增加、焦虑的情绪体验以及学习记忆能力的下降等。3.LBP在模型构建的过程中对脑力疲劳型心理亚健康模型大鼠的一般情况、自主活动情况、焦虑紧张情绪以及学习记忆下降等问题具有一定程度的预防和改善作用。
