炎症微环境下miR-140-5p靶向作用Smad3影响软骨细胞生物学特性的机制研究

颞下颌关节(Temporomandibular joint,TMJ)是颌面部唯一同时具备转动和滑动运动的左右联动关节,参与了咀嚼、吞咽、言语、表情和呼吸等精细且复杂的功能活动。骨关节炎(Osteoarthritis,OA)是关节疾病中的典型代表,其发病率高,对人体危害大,严重影响了患者的生活质量。虽有不少学者对这类关节疾病进行了相关研究报道,但目前对骨关节炎,尤其是颞下颌关节骨关节炎(Temporomandibular joint osteoarthritis,TMJ-OA)复杂的病理机制尚缺乏完整认识。以往的研究表明,miR-140-5p在软骨细胞分化和软骨内稳态过程中发挥着重要的作用,miR-140-5p基因敲除鼠会出现增龄性骨关节炎的症状。然而,临床研究却发现骨关节炎患者的关节软骨中miR-140-5p表达显著增加。虽然越来越多的研究认为miR-140-5p与OA关系密切,但其功能作用至今仍存在争议。有关miR-140-5p在OA,尤其在TMJ-OA中的作用机制亟需被阐明。本研究第一部分通过建立炎症微环境体外实验模型,模拟了 TMJ-OA样病损改变。第二部分探索了 miR-140...
