翼状胬肉的体外细胞模型建立和分化诱导治疗研究

目的:翼状胬肉是一种侵袭性增生,是角膜上皮细胞过度增殖的结果。分化治疗近年来引起了人们的广泛关注。本研究的目的是从翼状胬肉中鉴定干细胞,并检测翼状胬肉干细胞的分化策略。分化疗法的概念来源于激素、细胞因子或不同分化剂可以促进分化,并且可以不可逆地改变细胞表型。本文简要地介绍了几篇文献和研究,以证明目前的目标,即分化治疗在治疗多种临床疾病中能够发挥关键和有前途的作用。方法:应用体外原代培养技术获得胬肉干细胞,应用免疫荧光染色法研究培养的翼状胬肉干细胞的干细胞标志物和多向分化潜能等干细胞特性。诱导分化后利用克隆形成实验和MTT法检测细胞活力,检测和评价分化治疗策略在胬肉治疗中的应用潜能。结果:翼状胬肉干细胞可在体外进行持续的培养和扩增。它们表达了多能干细胞标记物,可在体外诱导分化为神经元细胞、骨细胞和脂肪细胞。经诱导试验处理后,翼状胬肉干细胞球表达干细胞标志物和上皮-间充质转化标志物。诱导分化后,翼状胬肉干细胞的增殖和存活能力受到抑制,提示分化诱导策略在翼状胬肉治疗中有广阔的应用前景。结论:翼状胬肉来源的细胞表达多种干细胞标志物并且具备多向诱导分化潜能。胬肉干细胞在体外经过诱导培养基处理后,...
