长白山区五种百合属植物保护研究

【摘要】:百合具有显著的经济价值、文化价值和药用价值。本文以长白山区五种百合属植物为试材开展研究。研究取得了以下结果:1.长白山区百合属五种植物在物种水平上均具有较高的遗传多样性,其中垂花百合的遗传多样性测度Shannon’s信息指数I=0.4393,毛百合I=0.4371、轮叶百合I=0.3975、细叶百合I=0.3948,珠芽百合I=0.3037。居群水平的遗传多样性研究结果:轮叶百合居群水平上的遗传多样明显低于物种水平,Shannon’s信息指数最高为0.2031,居群间遗传分化极大,基因流微弱(Gst=0.5248,Nm*=0.4527)。居群间和居群内的分化极为显著(P0.0010),Mantel分析表明遗传距离与水平地理距离具有显著的正相关性(r=0.0182,P0.01),遗传漂变是导致群体遗传结构变异的主要原因,而Φ值=0.581,表明轮叶百合居群以自交为优势,这种繁殖方式进一步加速了居群的分化,弱化了基因交流;毛百合居群水平上的遗传多样低于物种水平的遗传多样性,Shannon’s信息指数最高为0.3133,居群间遗传分化极大,基因流虽弱但大于1(Gst=0.3053,Nm*=1.1379),可以抵制遗传漂变对群体遗传结构的影响。居群间和居群内的分化极为显著(P0.0010),Mantel分析表明遗传距离与水平地理距离没有明显的相关性(r=-.7002,P0.05),而Φ值=0.417,表明毛百合居群兼有自交和杂交,这种繁殖方式削弱了居群的分化。细叶百合、垂花百合及珠芽百合的自然资源较少,仅有一个居群,无法进行居群间的比较分析。2.以ITS2通用引物为条形码完成了细叶百合的分子鉴定,同时我们还发现并使用3条ISSR引物(UBC807、UBC826、UBC866)实现了长白山区百合属5种百合的物种鉴别。3.轮叶百合与毛百合蛋白质含量低(分别为0.99%,0.44%,食用百合参考标准为6.7%),两种百合水溶性碳水化合物含量较高(分别为69.65%和76.18%,食用百合参考标准为79.5%)。冷浸出物含量较高(分别为37.93%,32.36%,中国药典要求≥18%),进一步利用HPLC法分析其药用成分甾体皂苷类成分以及粗多糖组分,发现:两种百合的甾体皂苷类成分含量较高,并且轮叶百合的甾体皂苷类成分高于毛百合,但粗多糖组分含量均较低。综上所述,细叶百合、垂花百合及珠芽百合在长白山区为稀缺自然资源,应采取相应的保护措施;轮叶百合及毛百合均有一定的药用开发价值,其种质资源的保护意义明显。
[Abstract]:Lily has remarkable economic value, cultural value and medicinal value. In this paper, five species of Lilium in Changbai Mountain area were used as experimental materials. The results of the study are as follows: 1: 1. The five species of Lilium in Changbai Mountain area all have high genetic diversity at species level. The genetic diversity measure of Lilium lilium is I ~ (0.4393), I ~ (0.3771), I _ (0.3975), I _ (0.3948), I _ (0.303737). The genetic diversity at population level was significantly lower than that at species level. The highest information index of Shannons was 0.2031. The genetic differentiation between populations was great and the gene flow was weak (Gst=0.5248,Nm*=0.4527). Mantel analysis showed that genetic distance was positively correlated with horizontal geographic distance (r = 0.0182, P0.01), and genetic drift was the main cause of genetic variation. The 桅 value of 0.581indicates that the population of Lilium rotundii is predominated by self-crossing, which further accelerates population differentiation and weakens gene exchange. The genetic diversity at population level of Lilium mongolica was lower than that at species level. The highest information index of Shannons was 0.3133.The genetic differentiation between populations was very large, and the gene flow was weak but greater than 1 (Gst=0.3053,Nm*=1.1379), which could resist the effect of genetic drift on the genetic structure of the population. The differentiation between populations and populations was extremely significant (P0.0010) Mantel analysis showed that there was no significant correlation between genetic distance and horizontal geographic distance (r-.7002p0.05), but 桅 value was 0.417, which indicated that the population of Lilium mongolica had both self-crossing and crossbreeding, which weakened the differentiation of population. The natural resources of Lilium lilium, Lilium sanguinalis and Lilium globosa are less, and there is only one population, which can not be compared and analyzed between populations. 2. The molecular identification of Lilium lilium was carried out by using ITS2 universal primers as bar code. At the same time, three ISSR primers (UBC807,UBC826,UBC866) were used to identify 5 species of Lilium in Changbai Mountain area. The content of protein was low (0.99% 0.44%, respectively, and the reference standard of edible lily was 6.7%), and the content of water-soluble carbohydrate was higher (69.65% and 76.18%, respectively). The reference standard of edible lily was 79.5%. The content of cold extract was higher (37.93% and 32.36%, respectively, and the requirement of Chinese pharmacopoeia was 鈮,
