组织处理对形态定量研究的影响

【摘要】: 第一部分放大倍数和包埋介质对睾丸组织形态定量研究的影响 研究背景与目的: 形态定量研究中,我们通常利用组织切片,根据体视学原理测量切片上观察到的图像,由此估计组织结构的形态定量特征。由于切片受取材、固定、脱水、包埋、切片、裱片和染色以及显微分辨率和放大倍数等诸多因素的影响,不同条件下测得的结果可能不同。这是一个基本而又重要的问题,但可能是因为影响因素(包括测量方法)太多,对这个问题的系统研究或综述相对很少。笔者实验室以前对比过国产石蜡与环氧树脂包埋对睾丸组织形态定量研究的影响,最近又对比了国际上常用的Leica石蜡和甲基丙烯酸树脂包埋以及放大倍数对肾脏组织形态定量研究的影响。本文拟进一步对比Leica石蜡和甲基丙烯酸树脂包埋以及不同物镜(不同分辨率和放大倍数)对睾丸组织结构形态定量研究的影响。 方法: 动物:6只正常成年(8.6~9.6月龄)雄性新西兰大耳白兔。麻醉后取每只兔的一侧睾丸(3只取自左侧,3只取自右侧)。经Bouin液
[Abstract]:Part one: the influence of magnification and embedding medium on the quantitative study of testicular tissue morphology: background and objective: In quantitative morphological studies, We usually use tissue sections to measure the images observed on the slices according to the stereology principle, and then estimate the morphological and quantitative characteristics of the tissue structure. Due to the influence of many factors, such as material selection, fixation, dehydration, embedding, slicing, laminating and dyeing, microresolution and magnification, the results may be different under different conditions. This is a basic and important problem, but it may be because there are so many influential factors (including measurement methods) that there is relatively little systematic study or review of the problem. The authors have compared the effects of paraffin wax and epoxy resin embedding on the quantitative study of testicular morphology in our laboratory. The effects of Leica paraffin wax and methacrylic resin embedding and magnification on the quantitative study of renal tissue morphology were also compared recently. The effects of Leica paraffin wax and methacrylic resin embedding and different objective lenses (different resolution and magnification) on the quantitative study of testicular tissue morphology were further compared in this paper. Methods: six normal adult (8.6 ~ 9.6 months old) male New Zealand white rabbits were used.
