TLR2和TLR4在结核分枝杆菌诱导肾小管上皮细胞损伤中的作用

【摘要】：目的：探讨Toll样受体2（TLR2）和Toll样受体4（TLR4）在牛型结核杆菌卡介苗（BCG）诱导人肾小管上皮细胞损伤中的作用。方法：用BCG刺激HK-2细胞，通过荧光定量PCR或Western blot方法分别检测TLR2、TLR4、转化生长因子-β1（TGF-β1）、趋化因子CX3CL1、E-钙粘蛋白（E-cad）和α-平滑肌肌动蛋白（α-SMA）的表达。将TLR2抗体以及TLR4拮抗剂CLI-90分别与BCG共刺激HK-2细胞，通过荧光定量PCR或Western blot方法分别检测TGF-β1、CX3CL1、E-cad和α-SMA的表达水平。结果：BCG刺激HK-2细胞可诱导TLR2、TLR4、TGF-β1、CX3CL1、α-SMA表达上调及E-cad表达下调。其中TLR2和TLR4的表达呈时间依赖性和剂量依赖性，TGF-β1、CX3CL1、α-SMA和E-cad表达则表现为时间依赖性。TLR2抗体和TLR4拮抗剂可部分抑制BCG引起的TGF-β1、CX3CL1、E-cad和α-SMA表达的变化。结论：BCG可通过激活TLR2和TLR4引起TGF-β1、CX3CL1、α-SMA表达上调及E-cad表达的下调，提示TLR2和TLR4介导的信号通路在结核分枝杆菌致肾小管上皮细胞损伤中发挥重要作用。
[Abstract]:Objective: to investigate the application of Toll like receptor 2 (TLR2) and Toll like receptor 4 (TLR4) in BCG vaccine of Mycobacterium bovis. Methods: BCG was used to stimulate HK-2 cells. TLR2, TLR4, TGF- 尾 1 and CX3CL1 were detected by fluorescence quantitative PCR or Western blot, respectively. E-cadA and 伪 -SMAA. TLR2 antibody and TLR4 antagonist CLI-90 were co-stimulated with BCG in HK-2 cells. TGF- 尾 1 CX3CL1 was detected by fluorescence quantitative PCR or Western blot. Results: TLR2TLR4TGF- 尾 1 and CX3CL1 could be induced in HK-2 cells stimulated by TLR2TLR4 and 伪 -SMA. The expression of 伪 -SMA was up-regulated and the expression of E-cad was down-regulated. The expressions of TLR2 and TLR4 were time-dependent and dose-dependent. The expression of 伪 -SMA and E-cad was time-dependent. TLR2 antibody and TLR4 antagonist could partially inhibit TGF- 尾 1 CX3CL1 induced by BCG. Conclusion the expression of TGF- 尾 1 and CX3CL1 can be induced by activation of TLR2 and TLR4. The up-regulation of 伪 -SMA and down-regulation of E-cad suggest that TLR2 and TLR4 mediated signaling pathway play an important role in renal tubular epithelial cell injury induced by Mycobacterium tuberculosis.
