2015年镇江地区832份腹泻样本诺如病毒感染状况及进化重组分析

【摘要】:目的分析2015年镇江地区诺如病毒的感染状况及进化重组现象。方法收集2015年镇江地区哨点医院腹泻患者的粪便标本及突发疫情的病例标本,采用实时荧光定量PCR(q RT-PCR)方法进行诺如病毒GI/GII核酸检测,阳性样本采用逆转录PCR(RT-PCR)方法扩增诺如病毒基因的Rd Rp区和Capsid区,序列测定、基因分型,运用Mega软件和RDP软件进行序列进化和基因重组分析。结果 832份样本中诺如病毒阳性率为5.05%(42/832),GII组阳性率为4.81%(40/832),占95.24%,GII组在10~20岁的中青年中阳性率最高,达25.00%(14/56)。2015年诺如病毒的检出率以1月最高,11月、2月次之,以GII.17型所占比例最高为63.89%(23/36),新GII.17型变异株为最主要的流行株。主要的重组事件为诺如病毒GII.P12_GII.3型的镇江毒株ZJ111/2015/CHN和ZJ119/2015/CHN,是由加拿大的参考株U02030/Toronto/CA/GII.3和英国的参考株X86557/Lordsdale/UK/GII.4重组而成。结论 2015年镇江地区诺如病毒感染以GII组为主,中青年阳性率最高,新GII.17型变异株为最主要的流行基因型,主要的重组事件为GII.3与GII.4的重组。
[Abstract]:Objective to analyze the infection status and evolutionary recombination of Norovirus in Zhenjiang area in 2015.Methods fecal specimens of diarrhea patients in Zhenjiang sentinel hospital in 2015 were collected, and real-time fluorescence quantitative PCR(q RT-PCR method was used to detect norovirus GI/GII nucleic acid.The Rd RP region and Capsid region of Norovirus gene were amplified by reverse transcription PCR RT-PCR method. Sequence determination, genotyping, sequence evolution and gene recombination analysis were performed by Mega software and RDP software.Results among 832 samples, the positive rate of norovirus was 5.05% / 832% in GII group (4.81% / 832%), accounting for 95.2420 years old, the highest positive rate was in young people aged 10 to 20 years, reaching 25.00% 14% 56%. In 2015, the positive rate of norovirus virus was highest in January, November and second month.The highest percentage of GII.17 type was 63.89%, and the new GII.17 variant was the main epidemic strain.The major recombination events were the Zhenjiang strain of Norovirus GII.P12_GII.3 ZJ111/2015/CHN and ZJ119 / 2015 / CHN, which were recombined from the reference strain U02030/Toronto/CA/GII.3 of Canada and the reference strain X86557/Lordsdale/UK/GII.4 of the United Kingdom.Conclusion the infection of Norovirus in Zhenjiang area in 2015 was mainly in GII group, the positive rate was the highest in young and middle-aged people, the new GII.17 variant was the main prevalent genotype, and the main recombinant event was the recombination of GII.3 and GII.4.
