新发展晚期血吸虫病患者血清肽谱分析

【摘要】:目的 :分析新发展晚期血吸虫病患者血清多肽,筛选有潜在应用价值的标志蛋白,为其诊断与发病机制研究奠定基础。方法:采集新发展晚期血吸虫病患者与健康人血清,利用包被有弱阳离子的磁珠富集纯化血清多肽,应用基质辅助激光解析电离时间飞行质谱(MALDI-TOF-MS)Flex分析技术结合Mascot生物信息学分析血清多肽指纹图谱。结果:与健康对照组比较,在0.8~13.0 ku区段,新发展晚期血吸虫病患者血清中m/z 2 082、4 282多肽表达上调(P0.01),2 661、2 991、3 241、5 337、5 906多肽表达下调(P0.05);常规晚期血吸虫病患者血清中m/z 2 662、4 209、5 337与5 906处显著下调,2 082处显著上调。结论:m/z3 241、4 282血清差异多肽对新发展晚期血吸虫病患者的诊断可能具有潜在应用价值,对该病发生发展的机制也奠定了理论基础。
[Abstract]:Objective: to analyze the serum polypeptides of patients with advanced schistosomiasis and to screen potential marker proteins for the diagnosis and pathogenesis of schistosomiasis. Methods: serum samples of newly developed advanced schistosomiasis patients and healthy persons were collected and purified by magnetic beads coated with weak cations. Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS Flex) was used to analyze serum polypeptide fingerprints with Mascot bioinformatics. Results: compared with the healthy control group, in the region of 0.8 ~ 13.0 ku, In the serum of newly developed advanced schistosomiasis patients, the expression of m / z 2 082n 4 282 polypeptide was up-regulated, and the expression of m- / z 2662 + 4209H 5 337 and 5 906 was significantly down-regulated at 2 082 and 2 082 points respectively in the serum of newly developed patients with advanced schistosomiasis, and the expression of P0.05906 polypeptide was down-regulated at 3241U 5337N 5906, and that in the serum of patients with advanced schistosomiasis was significantly lower than that in the patients with advanced schistosomiasis. ConclusionThe differential polypeptide in serum of w / m / z _ 3 241N _ 4 282 may have potential application value in the diagnosis of newly developed advanced schistosomiasis, and it also lays a theoretical foundation for the pathogenesis of schistosomiasis.
