不同浓度泡球蚴囊液干预对大鼠肝星状细胞TGF-β1、IL-6、TNF-α表达的影响

【摘要】:目的:研究泡球蚴囊液刺激环境对大鼠肝星状细胞(HSC-T6)TGF-β1、IL-6、TNF-α表达的影响。 方法:用浓度分别为0、0.01、0.025、0.05、0.1、0.2、0.4、0.9、1.7、3.4、6.8、13.5mg/ml的泡球蚴囊液与大鼠HSC-T6细胞共培养24h,,0mg/ml浓度组做空白对照。ELISA法(酶联免疫吸附测定法)检测各浓度泡球蚴囊液处理组转化生长因子β1(TGF-β1)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达变化。 结果:TGF-β1的表达在泡球蚴囊液浓度为0.01~6.8mg/ml范围内略高于空白对照组,但差异无统计学意义;泡球蚴囊液浓度高于3.4mg/ml时,IL-6的表达极显著增加(P 0.01),其中6.8、13.5mg/ml泡球蚴囊液处理组相比空白对照组分别增加3028.65、3879.23倍;泡球蚴囊液浓度≤0.2mg/ml时,TNF-α的表达趋于平稳;此后急剧增加又逐渐下降,其中泡球蚴囊液浓度为0.4mg/ml时TNF-α的表达量达到最大,为空白对照组的3.53倍,差异具统计学意义(P0.05)。 结论:泡球蚴囊液对大鼠HSC-T6细胞TGFβ1无明显的刺激分泌作用;高浓度的泡球蚴囊液可刺激大鼠HSC-T6细胞IL-6的表达分泌;一定浓度的泡球蚴囊液可刺激HSC-T6细胞TNF-α分泌。
