不同浓度泡球蚴囊液干预对大鼠肝星状细胞TGF-β1、IL-6、TNF-α表达的影响

【摘要】:目的:研究泡球蚴囊液刺激环境对大鼠肝星状细胞(HSC-T6)TGF-β1、IL-6、TNF-α表达的影响。 方法:用浓度分别为0、0.01、0.025、0.05、0.1、0.2、0.4、0.9、1.7、3.4、6.8、13.5mg/ml的泡球蚴囊液与大鼠HSC-T6细胞共培养24h,0mg/ml浓度组做空白对照。ELISA法(酶联免疫吸附测定法)检测各浓度泡球蚴囊液处理组转化生长因子β1(TGF-β1)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达变化。 结果:TGF-β1的表达在泡球蚴囊液浓度为0.01~6.8mg/ml范围内略高于空白对照组,但差异无统计学意义;泡球蚴囊液浓度高于3.4mg/ml时,IL-6的表达极显著增加(P 0.01),其中6.8、13.5mg/ml泡球蚴囊液处理组相比空白对照组分别增加3028.65、3879.23倍;泡球蚴囊液浓度≤0.2mg/ml时,TNF-α的表达趋于平稳;此后急剧增加又逐渐下降,,其中泡球蚴囊液浓度为0.4mg/ml时TNF-α的表达量达到最大,为空白对照组的3.53倍,差异具统计学意义(P0.05)。 结论:泡球蚴囊液对大鼠HSC-T6细胞TGFβ1无明显的刺激分泌作用;高浓度的泡球蚴囊液可刺激大鼠HSC-T6细胞IL-6的表达分泌;一定浓度的泡球蚴囊液可刺激HSC-T6细胞TNF-α分泌。
[Abstract]:Objective: To study the effect of echinococcus cyst fluid stimulation on the expression of TGF- beta 1, IL-6 and TNF- alpha in rat hepatic stellate cells (HSC-T6).
Methods: the alveolar cyst fluid with the concentration of 0,0.01,0.025,0.05,0.1,0.2,0.4,0.9,1.7,3.4,6.8,13.5mg/ml and the rat HSC-T6 cells were co cultured with 24h, and the concentration group of 0mg/ml was used as the blank control.ELISA (enzyme linked immunosorbent assay) to detect the transforming growth factor beta 1 (TGF- beta 1), interleukin 6 (IL-6), and the swelling of interleukin 6 (IL-6) in the treatment group. The changes in the expression of tumor necrosis factor - alpha (TNF- - alpha).
Results: the expression of TGF- beta 1 was slightly higher than that of the blank control group in the range of 0.01 ~ 6.8mg/ml of alveolar cyst fluid, but the difference was not statistically significant. The expression of IL-6 was significantly increased (P 0.01) when the concentration of vesicle cyst fluid was higher than that of 3.4mg/ml. The 6.8,13.5mg/ml alveolar cyst fluid was increased by 3028.653879.23, respectively, compared with the blank control group. When the concentration of vesicle cyst fluid was less than 0.2mg/ml, the expression of TNF- alpha tended to be stable and then increased rapidly and gradually, and the expression of TNF- a was the maximum when the concentration of vesicle cyst fluid was 0.4mg/ml, which was 3.53 times that of the blank control group, and the difference was statistically significant (P0.05).
Conclusion: the hydatid cyst fluid has no obvious stimulating effect on TGF beta 1 of rat HSC-T6 cells, and the high concentration of alveolar cyst fluid can stimulate the expression and secretion of IL-6 in rat HSC-T6 cells, and a certain concentration of vesicle sac can stimulate the secretion of TNF- alpha in HSC-T6 cells.
