不同滴度噬菌体D29对气道上皮细胞9HTE的生长及功能影响研究

【摘要】:目的研究噬菌体D29不同滴度对气道上皮细胞株9HTE的生长及功能影响。方法分别用高滴度(109PFU/mL)和低滴度(107 PFU/mL)噬菌体D29及噬菌体缓冲液组作用于9HTE细胞株后,用四甲基偶氮唑蓝比色法(MTT)检测细胞生长情况;用酶联免疫吸附试验(ELISA)法测量细胞上清液白细胞介素(IL)-6、IL-8水平;用逆转录PCR(RT-PCR)检测细胞间黏附分子-1(ICAM-1)mRNA的表达,用流式细胞技术检测细胞凋亡率。结果高、低滴度噬菌体D29作用气道上皮细胞一段时间后,对细胞生长活性、细胞因子IL-6、IL-8和ICAM-1mRNA的表达,以及细胞凋亡率的影响与噬菌体缓冲液组比较差异无统计学意义(P0.05)。结论高、低滴度噬菌体D29对体外培养的气道上皮细胞生长及功能无明显影响。
[Abstract]:Objective to study the effects of different titers of phage D 29 on the growth and function of airway epithelial cell line 9 HTE. Methods High titer (109 PFU / mL) and low titer (107 PFU / mL) phage D29 and bacteriophage buffer group were used to treat 9HTE cell line respectively. The cell growth was detected by MTT assay. The levels of interleukin-6 (IL-8) in supernatant were measured by enzyme-linked immunosorbent assay (Elisa), the expression of intercellular adhesion molecule-1 (ICAM-1) mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR), and the apoptosis rate was detected by flow cytometry. Results the effects of high and low titer phage D29 on the cell growth activity, the expression of cytokines IL-6, IL-8 and ICAM-1 mRNA, and the rate of apoptosis were not significantly different from those of phage buffer group (P0.05). Conclusion High and low titer phage D 29 has no effect on the growth and function of cultured airway epithelial cells.
