陕西省部分地区人、畜禽戊型肝炎病毒抗体的检测

【摘要】:戊型肝炎病毒是戊型肝炎的病原体,可引起急性、病毒性肝炎,表现为腹痛、呕吐、发热、厌食、恶心、乏力、并伴有黄疸、尿色深和肝脾肿大,重度肝炎可表现为肝昏迷、鼻出血、消化道出血、皮肤淤斑、急性脑水肿等。患者及隐性感染者是该病的主要传染源。HEV主要通过粪-口途径传播,多集中在发展中国家,在美国、法国、日本、希腊、西班牙、意大利等发达国家也有越来越多关于戊型肝炎的报道。而我国有2.1亿人感染了HEV。因此,戊型肝炎已成为重要的公共健康问题。 禽戊型肝炎病毒是鸡的肝炎脾大或大肝大脾病综合征的主要病原,在世界许多国家广泛流行,严重影响了养禽业的健康发展。为了调查禽戊型肝炎病毒中国株(CaHEV)抗体在不同动物中的分布情况,,本试验利用间接酶联免疫吸附法(ELISA),将原核表达的CaHEV ORF2羧基端268个氨基酸编码的重组蛋白和禽戊型肝炎病毒特有的抗原表位模拟肽peptide-2分别作为包被抗原,对陕西省部分地区人、猪、鸡、犬、牛、羊血清和牛奶、羊奶的抗禽HEV ORF2蛋白的抗体进行了检测。结果表明,人、猪、鸡、犬血清和牛奶、羊奶中抗禽HEV ORF2的抗体阳性率分别为10.32%(67/1677)、3.57%(1/28)、2.9%(1/34)、61.6%(53/86)和3.39%(2/58)、0.56%(1/177),用禽HEV特有的抗原表位peptide-2检测发现,上述阳性样品中含有特异性抗禽HEV抗体的阳性率为8.95%(6/67)、0(0/1)、0(0/1)、28.3%(15/53)和50%(1/2)、100%(1/1),牛、羊血清中未发现阳性样本。 结果提示,人、猪、鸡、犬、牛和羊都有禽HEV特异性抗体。其中犬的阳性率最高,但对含有禽HEV特异性抗体的血清样本进行RT-nPCR扩增禽HEV RNA,没有扩增到目的基因。因此,并不能证明犬感染了禽HEV。本研究通过间接ELISA法对陕西省部分地区人畜中禽戊型肝炎病毒抗体进行了检测,丰富了禽戊型肝炎病毒的流行病学资料,为研究禽戊型肝炎病毒的致病性及可能的人畜共患性奠定基础。
[Abstract]:Hepatitis E virus is the pathogen of hepatitis E, which can cause acute and viral hepatitis. It is characterized by abdominal pain, vomiting, fever, anorexia, nausea, fatigue, accompanied by jaundice, urine color and hepatosplenomegaly. Epistaxis, gastrointestinal bleeding, skin stasis, acute brain edema and so on. Patients and recessive infections are the main source of infection of the disease. HEV is mainly transmitted through faecal-oral routes, mostly concentrated in developing countries, in the United States, France, Japan, Greece, Spain, More and more reports of hepatitis E have been reported in developed countries such as Italy. And 210 million people in China are infected with HEV.. Therefore, hepatitis E has become an important public health problem. Avian hepatitis E virus is the main pathogen of hepatitis spleen or large liver and spleen disease syndrome in chicken. It is widely prevalent in many countries all over the world, which seriously affects the healthy development of poultry industry. In order to investigate the distribution of (CaHEV) antibody against avian hepatitis E virus Chinese strain in different animals, In this study, indirect enzyme-linked immunosorbent assay (ELISA),) was used to encode the recombinant protein encoding 268 amino acids at the carboxyl end of CaHEV ORF2 and the epitope mimic peptide peptide-2 of avian hepatitis E virus as coating antigens. Antibodies against avian HEV ORF2 protein were detected in pig, chicken, dog, cattle, sheep serum and milk and goat milk. The results showed that the positive rates of anti-avian HEV ORF2 in human, pig, chicken, dog serum, milk and goat's milk were 10.32% (67 / 1677), 3.57% (1 / 28), 2.9% (1 / 34), 61.6% (53 / 86) and 3.39% (2 / 58) 0.56% (1 / 177), respectively. The positive rate of the above positive samples was 8.95% (6 / 67) 0 (0 / 1) 0 (0 / 1) or 28.3% (15 / 53) and 50% (1 / 2) 100% (1 / 1) respectively. No positive samples were found in serum of cattle and sheep. The results suggest that human, pig, chicken, dog, cattle and sheep have avian HEV specific antibodies. The positive rate was the highest in dogs, but the target gene was not amplified by RT-nPCR in serum samples containing specific antibodies to avian HEV. Therefore, it does not prove that the dog is infected with avian HEV. In this study, indirect ELISA method was used to detect avian hepatitis E virus antibody in human and livestock in Shaanxi Province, which enriched the epidemiological data of avian hepatitis E virus. To lay a foundation for studying the pathogenicity and possible zoonosis of avian hepatitis E virus.
