TNF-α基因多态性与青海地区藏族人群乙肝病毒易感性的相关性研究
[Abstract]:Objective: to analyze the relationship between the susceptibility of chronic hepatitis B (CHB) and the two loci of TNF- 伪 gene-238 and -308 in Tibetan population in Qinghai area, and to screen out the main related loci. To lay a molecular foundation for the prediction of HBV susceptibility risk in Qinghai Tibetan population. So as to provide new ideas and methods for clinical diagnosis and prevention of chronic hepatitis B. Methods: (1) study subjects: 250 Tibetan chronic hepatitis B patients (case group) and 134 healthy persons (control group) in Qinghai. (2) Polymorphism Analysis of Gene: peripheral DNA, was extracted from peripheral blood by using a small quantity extraction kit of blood genome. The TNF- 伪 gene locus was analyzed by FMS to determine -238A / G / -238A / An / 238G / A / G / 308G / A, -genotype of 308G/G. (3) data analysis: the frequencies of genotypes and alleles were calculated, the differences of genotypes were compared, and the Hardy-Weinberg equilibrium coincidence test was performed. The correlation between genotypes and susceptibility to hepatitis B was analyzed by logistic multivariate regression analysis. Results: (1) the frequency distribution of -238 locus in the case group and the control group:-238A/G was 8.4% and 8.2% in the case group and the control group, respectively. There was no significant difference in distribution frequency among groups. -238A/A in the case group and the control group were 0.4% and 0.0.The results of statistical analysis showed that there was no significant difference in the distribution frequency among the groups. -238G/G was 91.2% and 91.8% in the case group and the control group, respectively. The results of statistical analysis showed that there was no significant difference in the distribution frequency among the three groups. (2) the distribution of -308 in each clinical type:-308G/A in chronic toxic hepatitis B and normal control group were 7.6% and 7.5%, respectively. -308G/G was 92.4% and 92.5% in each group, and there was no significant statistical difference in the distribution frequency between each group. Conclusion: (1) the polymorphism of TNF- 伪 -238 gene may not be associated with the susceptibility to HBV infection in Tibetan population in Qinghai; (2) the polymorphism of TNF- 伪 -308 gene may not be associated with the susceptibility to HBV infection in Tibetan population in Qinghai.
