日本血吸虫尾蚴逸出节律性及尾蚴趋向吸附的研究

【摘要】:第一部分日本血吸虫尾蚴逸出昼夜节律性及光照周期变化对其影响 目的:观察感染性钉螺体内日本血吸虫尾蚴逸出的昼夜节律性,以及光照周期变化对此特征的影响。 方法:以安徽(山区)和江苏(湖区)两地感染性钉螺为研究对象,分别将钉螺置于自然光照、昼夜颠倒、7:00~9:00插入短暂黑暗、17:00~19:00插入短暂黑暗这四种光照周期条件下连续逸蚴24h,记录每2h时间段内感染性钉螺体内日本血吸虫尾蚴逸出数量,,从而探讨光照周期变化与尾蚴逸出的关系。 结果:在自然光照周期下,山区感染性钉螺体内日本血吸虫尾蚴逸出高峰时段为下午17:00~21:00,表现为‘晚逸蚴’;湖区感染性钉螺体内日本血吸虫尾蚴逸出高峰时段为上午7:00~11:00,表现为‘早逸蚴’。在昼夜颠倒的光照周期下,湖区感染性钉螺体内日本血吸虫尾蚴逸出节律性出现逆转;上午7:00~9:00插入短暂黑暗会相应延迟山区和湖区感染性钉螺体内日本血吸虫尾蚴逸出高峰时段;下午17:00~19:00插入短暂黑暗对两地感染性钉螺体内日本血吸虫尾蚴逸出节律性几乎没有影响。 结论:日本血吸虫尾蚴逸出呈现昼夜节律性,光照周期变化会相应改变尾蚴逸出节律性。 第二部分日本血吸虫尾蚴的趋向吸附作用以及温度和尾蚴时龄对其影响 目的:观察日本血吸虫尾蚴对油酸的趋向吸附能力,以及尾蚴时龄和水温对此特征的影响。 方法:利用蜂蜡、指甲油的可凝固性与不同浓度(0.15g/ml、0.3g/ml、0.6g/ml)油酸混合后制备成一种皮肤模拟物,供尾蚴吸附。空白组、蜂蜡组、指甲油组作为对照组。模拟小鼠腹部贴片感染尾蚴的方法,用接种环挑取50±5条尾蚴,置于预先制备好的皮肤模拟物上,一段时间后滴加卢戈氏碘液固定染色,显微镜下计算皮肤模拟物上尾蚴断尾率。或揭去盖玻片,用去氯水多次冲洗皮肤模拟物后滴加卢戈氏碘液固定染色,显微镜下计数经去氯水冲洗后仍吸附在皮肤模拟物上的尾蚴数量。 结果:油酸组皮肤模拟物上尾蚴断尾率及吸附数量与对照组相比较存在极其显著性差异(P<0.01)。尾蚴与皮肤模拟物接触时间相等时,0.15g/ml油酸组与0.3g/ml油酸组皮肤模拟物上尾蚴断尾率存在显著性差异(P<0.05),0.3g/ml油酸组与0.6g/ml油酸组皮肤模拟物上尾蚴断尾率不存在显著性差异。油酸浓度相同时,逸出后0h尾蚴与逸出后5h尾蚴在皮肤模拟物上吸附数量无显著性差异;逸出后5h尾蚴与逸出后10h尾蚴在皮肤模拟物上吸附数量存在显著性差异(P<0.05)。水温变化(25~35℃)对尾蚴断尾率及吸附数量均无显著性差异。 结论:日本血吸虫尾蚴对油酸具有趋向吸附作用,尾蚴时龄影响尾蚴趋向吸附的能力。水温变化(25~35℃)对其影响不大。
