【摘要】:本文旨在通过体外模拟瘤胃法联合体内验证试验综合评定筛选出酵母培养物(Yeast Culture,YC)对瘤胃发酵具有最佳效果的适宜添加量,并用该酵母培养物其最佳添加量与达农威的益康XP比较,评价两种酵母培养物对绵羊瘤胃发酵及消化道内营养物质流通与消化的影响的效果异同。本研究采用9只身体状况良好、体重相近(35±2.67 kg),并装有永久性瘤胃瘘管、十二指肠瘘管和回肠瘘管的小尾寒羊半同胞公羊作为实验动物。论文共分为三个部分:首先,通过体外及体内试验,对本试验所用YC与达农威益康XP进行比较,筛选出在绵羊日粮精粗比3:7条件下本试验YC的适宜添加量。采用两因素交叉分组试验设计,试验分为两期:一期为体外试验,试验分为5组,对照组为不添加YC的基础日粮,实验II、III、IV组在基础日粮上添加酵母培养物的剂量分别为1%、1.5%、2%,第V组在基础日粮上添加2%的益康XP,每组三个重复,依据试验结果筛选出最佳的两组添加剂量;二期为体内试验,试验分为三组:对照组,体外筛选结果最佳的前两个添加剂量组,每组三个重复,体外及体内均测定3个时间段的pH值、NH3-N、VFA浓度。结果表明:(1)试验一:从整体趋势看,III组、IV组与V组发酵效果最接近,能显著降低NH3-N浓度、提高乙酸、丙酸、丁酸浓度(P0.05)。(2)试验二:III组能显著提高TVFA浓度、降低NH3-N浓度(P0.05),确定III组为绵羊的适宜添加量。其次,比较本试验YC与益康XP对绵羊瘤胃发酵的影响。按试验要求随机分为3组,每组3个重复,I组为对照组,II组投喂1.5%YC,III组投喂2%益康XP。结果表明:II组、III组与I组比较均能显著提高pH值(P0.05)。II组NH3-N浓度显著低与I组(P0.05),III组NH3-N浓度极显著低于I组(P0.01),显著低于II组(P0.05)。II组、III组均能极显著提高总酸、乙酸、丙酸浓度(P0.01)。结果表明:添加1.5%酵母培养物能够与添加2%益康XP对稳定采食后瘤胃pH值、降低瘤胃NH3-N浓度、提高瘤胃TVFA浓度具有一致的显著效果,并具有有提高瘤胃BCP浓度的趋势。最后,比较本试验YC与益康XP对绵羊消化道内营养物质流通与消化的影响。按试验要求随机分为3组,每组3个重复,I组为对照组,II组投喂1.5%YC,III组投喂2%益康XP。结果表明:(1)两种YC均降低了瘤胃液相流通速率、瘤胃液相体积以及滞留时间和DM(干物质)、OM(有机物)在直肠的流通量(P0.05),益康XP降低了ADF(酸性洗涤纤维)在直肠的流通量(P0.05)。(2)两种YC均提高了瘤胃MN(微生物氮)流通量和十二指肠N(氮)的流通量,以及DM、OM、NDF(中性洗涤纤维)、ADF在全消化道的表观消化率(P0.05),YC提高了CP在小肠的表观消化率(P0.05)。结果表明,YC可能是通过提高营养物质在小肠的表观消化率,从而提高全消化道表观消化率。
[Abstract]:The aim of this study was to select the optimal dosage of yeast culture YC for rumen fermentation by in vitro mimic rumen method combined with in vivo validation test. The optimum addition amount of the yeast culture was compared with the Yikang XP of Danongwei. To evaluate the effects of two yeast cultures on rumen fermentation and nutrient circulation and digestion in the digestive tract of sheep, 9 of them were in good physical condition with similar body weight of 35 卤2.67 kg). The small tail Han sheep half-sibling ram with permanent rumen fistula duodenal fistula and ileal fistula were used as experimental animals. The thesis was divided into three parts: firstly the experiment was carried out in vitro and in vivo. By comparing YC with Danongwei Yikang XP, the suitable addition amount of YC was selected under the condition of 3: 7 forage ration of sheep diet. Two factors cross grouping experiment design was used. The experiment was divided into two periods: the first stage was in vitro trial, the experiment was divided into five groups, the control group was the basal diet without YC, and the experiment was IIHIII. The dosage of yeast culture added to the basal diet in group IV was 1: 1. 5 and 2 / 2 respectively. Group V added 2% of Yikang XP to the basal diet, which was repeated three times in each group. According to the test results, the best two groups of additives were selected. The second phase was in vivo trial. The experiment was divided into three groups: control group, the first two dosage groups with the best screening results in vitro, each group with three repetitions. The pH value of NH3-N was measured in vitro and in vivo. VFA concentration. The results showed that: 1. According to the overall trend, the fermentative effect of group IV and group V was the closest, which could significantly reduce the concentration of NH3-N and increase acetic acid and propionic acid. The concentration of butyric acid in group 2: III could significantly increase the concentration of TVFA and decrease the concentration of NH3-N (P0.05). Then, the effects of YC and Yikang XP on rumen fermentation of sheep were compared. According to the requirements of the experiment, the III group was randomly divided into 3 groups, each group with 3 repeats as the control group. Group II was fed 1.5yycyou III group and fed with 2% Yikang XP.The results showed that group II was fed with 1. 5% YCX III. Compared with group I, the concentration of NH3-N in III group was significantly higher than that in group I (P 0.05). The concentration of NH3-N in group II was significantly lower than that in group I (P 0.05). The concentration of NH3-N in group III was significantly lower than that in group I (P 0.01) and significantly lower than that in group II (P 0.05). Group II could significantly increase total acid and acetic acid. The results showed that 1.5% yeast culture and 2% Yikang XP could decrease the rumen pH value and reduce the rumen NH3-N concentration after feeding steadily. The increase of rumen TVFA concentration has a consistent and significant effect, and has the trend of increasing rumen BCP concentration. Finally. The effects of YC and Yikang XP on nutrient circulation and digestion in digestive tract of sheep were compared. III group was fed with 2% Yikang XP.The results showed that both YC and YC decreased rumen liquid flow rate, rumen liquid volume, retention time and DM (dry matter). Om (organic matter) flux in the rectum (P0.05). Yikang XP decreased the flux of ADF (acid detergent fiber) in the rectum (P0.05. 2) both YC and YC increased the flux of MN (microbial nitrogen) and duodenum (N) in the rumen. The flux. And the apparent digestibility of ADF in the whole digestive tract (P0.05). YC increased the apparent digestibility of CP in the small intestine. The results showed that YC could improve the apparent digestibility of the whole digestive tract by increasing the apparent digestibility of nutrients in the small intestine.
