【摘要】:绵羊脂肪沉积的部位和沉积方式显著影响羊肉品质和养羊效益。因此,揭示功能基因、日粮及生长阶段对绵羊脂肪沉积的影响,对改良绵羊肉品质具有重要意义。研究选取了脂肪代谢相关功能基因NELL1和FMO3,比较其在不同尾型绵羊品种(哈萨克羊、滩羊、同羊、西藏羊)尾部脂肪组织中的表达差异;探讨营养水平及生长阶段对其在滩羊脂肪组织中表达的影响,旨在揭示不同营养水平及不同生长阶段脂尾型绵羊NELL1和FMO3基因表达的组织特异性及其表达模式,为脂尾型绵羊体内脂肪沉积的营养调控提供理论依据。试验一:不同品种绵羊尾脂中NELL1和FMO3 mRNA表达差异分析选择健康、体重相近的9月龄滩羊、同羊、哈萨克羊和西藏羊羯羊各3只,采集尾部脂肪组织,利用实时荧光定量PCR技术,检测NELL1和FMO3 mRNA表达,结果如下:1.FMO3mRNA在哈萨克羊尾脂中表达量最高(P0.05);在同羊、滩羊、西藏羊尾脂中依次下降,且差异显著(P0.05)。2.NELL1mRNA在西藏羊尾脂中表达量最高(P0.05);在滩羊、同羊、哈萨克羊中表达量依次下降。试验二:营养水平对不同阶段滩羊部分组织中NELL1和FMO3 mRNA表达的影响选择健康4月龄滩羊112只随机分为4组,公母各半,每组四个重复,每个重复7只羊。试验根据增重目标分为3个阶段:第一阶段(22~28kg),第二阶段(28~35kg),第三阶段(35~40kg),每个阶段各组分别饲喂不同营养水平的日粮,由低到高分为T1、T2、T3、T4组。每个生长阶段结束时采集肝脏、肾周脂肪、尾脂、皮下脂肪的组织样品,提取样品总RNA,采用实时荧光定量PCR的方法分析NELL1和FMO3 mRNA的表达差异。结果如下:1.NELL1 mRNA在尾脂中的表达,第1阶段以T4组为最高(P0.05),其他三组差异不显著;第2阶段和第3阶段,也都是以T4组表达量最高(P0.05),T1、T2、T3组表达量略有波动。2.NELL1 mRNA在皮下脂中的表达。第1阶段和第2阶段四个营养水平组间表达差异不显著(P0.05);第3阶段,T4组表达量最高(P0.05),依次为T2、T3、T1组。相同营养水平下,NELL1 mRNA在第1阶段表达量最高(P0.05)。3.NELL1 mRNA在肾周脂中的表达。在同一生长阶段,营养水平对滩羊肾周脂肪组织中NELL1mRNA的表达无显著影响(P0.05);在相同营养水平下,不同生长阶段间表现出表达差异显著(P0.05)。4.NELLL1 mRNA在肝脏中的表达。第1阶段,在四个营养水平组间表达差异不显著;第2阶段,表达以T4组最高(P0.05),依次为T3、T1、T2组;第3阶段,表达以T1组为最高(P0.05),依次为T2、T3、T4组。3个阶段表达依次升高,其中第3阶段表达量最高(P0.05)。5.FMO3 mRNA的表达。尾部脂肪组织中最高表达均出现在T2组;在第1阶段、第2阶段和第3阶段,T1、T2、T3、T4组中表达量均呈现先升高再降低的趋势;皮下脂肪组织T3组最高,肾周脂最高表达出现在T1组,肝脏的最高表达出现在第T4组,最高表达组与相同阶段时的最低表达组相比差异显著的(P0.05)。试验三:NELL1、FMO3mRNA在滩羊不同组织中的表达差异分析在试验第三阶段结束当天,选取T2组4只滩羊进行屠宰,采集肝脏、肾周脂肪、尾脂、皮下脂肪组织,利用实时荧光定量PCR技术,检测NELL1、FMO3mRNA的相对表达量,结果如下:1.NELL1 mRNA在滩羊尾部脂肪组织、皮下脂肪组织、肾周脂肪组织、肝脏中均有表达。NELL1在肝脏组织中表达最高,显著高于尾脂、皮下脂和肾周脂(P0.05),其次是在肾周脂中的表达,高于尾脂和皮下脂(P0.05),在尾脂和皮下脂中的表达最低,两者之间差异不显(P0.05)。2.FMO3 mRNA在滩羊尾部脂肪组织、皮下脂肪组织、肾周脂肪组织、肝脏中均有表达。FMO3在肝脏组织中表达最高,显著高于尾部脂肪组织、皮下脂肪组织和肾周脂肪组织(P0.05),其次是在尾部脂肪组织中的表达,高于肾周脂肪组织和皮下脂肪组织(P0.05),在肾周脂肪组织和皮下脂肪组织中的表达最低,两者之间差异不显(P0.05)。综合以上试验结果,表明NELL1和FMO3基因在绵羊组织中的表达受到品种、部位、生长阶段、营养水平等原因会有不同的表达结果,其表达差异是与脂肪的沉积差异是存在一定关系。
