PRRSV强弱毒体外诱导IL-10产生差异的分子基础研究

【摘要】:猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)感染后能够引发机体产生免疫抑制,IL-10在病毒介导的免疫抑制中起重要作用。PRRSV HuN4株感染PAM细胞后能够诱导IL-10高水平表达,而其传代致弱毒株HuN4-F112感染PAM细胞后IL-10表达水平很低。本研究以PRRSV HuN4株及HuN4-F112株为研究对象,对其感染PAM细胞后IL-10上游关键因子表达差异进行研究,揭示诱导IL-10产生差异的分子基础。结果显示,PRRSV强弱毒感染PAM细胞后,强毒HuN4株诱导TLR2与TLR4转录水平显著高于弱毒HuN4-F112株,TLR3、TLR7和TLR9转录水平则无显著差异;强毒HuN4株感染PAM细胞后,其p38磷酸化水平显著高于弱毒HuN4-F112株,而ERK磷酸化水平差异不具备显著统计学意义。结果提示,PRRSV强弱毒株感染PAM细胞诱导IL-10表达水平的差异推测是由于强弱毒诱导p38 MAPK磷酸化水平差异造成的。
[Abstract]:Porcine reproductive and respiratory syndrome virus. PRRSV-) infection can induce immune suppression in the body. IL-10 plays an important role in virus-mediated immunosuppression. PRRSv HuN4 strain can induce high IL-10 expression after infecting PAM cells. However, the expression of IL-10 was very low after the subculture of attenuated strain HuN4-F112 infected PAM cells. In this study, PRRSV HuN4 and HuN4-F112 strains were selected as the research objects. The differential expression of key factors in upstream of IL-10 was studied after its infection with PAM cells to reveal the molecular basis of inducing the difference in IL-10. After PRRSV was infected with PAM cells, the transcription levels of TLR2 and TLR4 induced by HuN4 strain were significantly higher than those of HuN4-F112 strain. There was no significant difference in transcription level between TLR7 and TLR9. The phosphorylation level of p38 in PAM cells infected with highly virulent HuN4 strain was significantly higher than that in attenuated HuN4-F112 strain, but there was no significant difference in ERK phosphorylation level. The difference of IL-10 expression induced by PAM cells infected with PRRSV strain is probably due to the difference of phosphorylation level of p38 MAPK induced by strong and weak virulence.
