羊源沙门氏菌耐药性和耐药基因的检测

【摘要】:为掌握新疆某养殖场羊源沙门氏菌对常用抗菌药物的耐药情况,并了解其携带部分相关耐药基因的流行现状及β-内酰胺酶、16SrRNA甲基化酶和喹诺酮类耐药基因(Plasmid mediated quinolone resistance,PMQR)的共存情况。使用琼脂稀释法对分离的沙门氏菌进行药敏试验,通过PCR方法进行blaTEM、blaCMY-2、blaCTX-M、blaLAP-1、blaKPC、blaOXA、blaSHV等β-内酰胺酶和armA、rmtB等16SrRNA甲基化酶及qnrA、qnrB、qnrC、qnrD、qnrS、qepA、oqxA、oqxB和aac(6′)-Ib-cr等PMQR基因检测,分析阳性菌株携带的基因型与耐药表型之间的关系。结果显示新疆该羊养殖场分离的沙门氏菌对被检的12种抗菌药物耐药率超过80%的有6种。47株菌携带blaTEM基因,34株菌携带blaOXA基因;未检出armA和rmtB等16SrRNA甲基化酶。33株菌携带qnrS,46株菌携带oqxA,46株菌携带oqxB,47株菌携带acc(6′)-Ib-cr基因。新疆该养殖羊场分离株耐药现象严重,携带耐药基因以blaTEM、blaOXA、qnrS、aac(6′)-Ib-cr、oqxA和oqxB为主,耐药基因型复杂多变、呈多样化发展,应该加强对β-内酰胺酶及PMQR因子的监控。
[Abstract]:In order to understand the drug resistance of sheep origin Salmonella to common antimicrobial agents in a Xinjiang farm, and to understand the prevalence of some related drug resistance genes and 尾 -lactamases. 16s rRNA methylase and Quinolone resistance gene Plasmid mediated quinolone resistance. The coexistence of PMQR. The susceptibility of isolated Salmonella was tested by Agar dilution method and blaTEMblaCMY-2 blaCTX-M was carried out by PCR method. The 尾 -lactamases, such as 尾 -lactamases and 16s rRNA methylases such as armAgnrmtB and qnrAnrB. The detection of PMQR genes such as qnrC, QnrDN, qnrS, qnrSN, qnrSN, qnrSN, QnrSN, QnrSN, QnrSN, QnrS@@. The relationship between the genotypes of positive strains and the phenotype of drug resistance was analyzed. The results showed that 6 of 47 strains of Salmonella isolated from the sheep farm in Xinjiang carried more than 80% antimicrobial resistance rates. With the blaTEM gene. 34 strains carried blaOXA gene. No 16s rRNA methylases, such as armA and rmtB, were detected. 33 strains of them carried oqxB. 47 strains of bacteria carried acc(6')-Ib-cr gene. The drug resistance of the isolates isolated from the sheep farm in Xinjiang was serious, and the drug resistance gene was blaTEM-OXAX qnrS. The drug resistant genotypes were complex and varied, so the monitoring of 尾 -lactamases and PMQR factors should be strengthened.
