捕食线虫性真菌Duddingtonia flagrans不同批量培养基的筛选与临床应用模式研究

【摘要】:Duddingtonia flagrans是目前发现的对动物线虫病生物防治最具有应用潜力的一种捕食线虫性真菌。为了探讨其批量培养的最佳方式以及在我国畜牧业生产实际中的临床应用模式,本研究通过比较D.flagrans的不同培养条件,筛选适用于生产实践的批量培养基和培养时间。同时,制备D.flagrans冻干生物制剂并进行与常用驱线虫药联合使用的试验,以探讨利用捕食线虫性真菌进行生物防治线虫感染的临床应用模式,为捕食线虫性真菌在生产实际中的规模化应用提供基本参考数据。研究结果表明:捕食线虫性真菌-D.flagrans在培养至第3-4周时,制备成冻干生物制剂,厚壁孢子复苏后的萌发率和菌丝生长率最佳,且玉米粒批量培养基与大麦粒批量培养基统计学差异不显著(P0.05),但前者更好一些。冻干生物制剂于4℃冰箱中保存半年以上,孢子的活力没有发生明显变化。捕食线虫性真菌-D.flagrans冻干生物制剂与伊维菌素(Ivermectin)联合应用于绵羊的寄生性线虫病防治,平均幼虫减少率可达到86.7%;与丙硫咪唑(Albendazole)联合应用幼虫减少率更高,为99.2%,而单独用菌组或单独用药组与上述真菌和驱虫药联合使用组相比,平均幼虫减少率差异显著(P0.05),说明捕食线虫性真菌-D.flagrans冻干生物制剂与驱虫药联合使用的临床应用模式,可以取得较好的寄生性线虫病临床防控效果,值得进一步在生产实践中进行深入研究和推广。
[Abstract]:Duddingtonia flagrans is one of the most promising nematode predatory fungi for biological control of animal nematode disease. In order to explore the best way of batch culture and clinical application in animal husbandry production in China. In this study, by comparing the different culture conditions of D. flagrans, the batch culture medium and culture time suitable for production practice were selected. Meanwhile, the freeze-dried biological preparations of D. flagrans were prepared and used in combination with common nematode repellent agents. To explore the clinical application model of biological control of nematode infection by nematode predatory fungi. The results showed that the nematode predator fungus -D.flagrans was prepared into freeze-dried biological agent at the 3-4 weeks of culture, and provided the basic reference data for the large-scale application of nematode predatory fungi in the production practice. The germination rate and hyphae growth rate of thick-walled spores were the best after resuscitation, and there was no significant difference between cornkernel batch culture medium and large wheat grain batch medium, but the former was better.
