秦川牛ELOVL6基因SNP检测及其与体尺、体重、胴体重性状的关联性分析

【摘要】:超长链脂肪酸延伸酶6是ELOVLs家族中的第6个成员,有5个外显子、4个内含子。£10叹6的功能主要表现在促成C1 6饱和与不饱和脂肪酸转变成为18C脂肪酸。ELOVLs家族的基因在动物机体内的控制和表达调控主要表现在脂肪,尤其是超长链。脂肪酸是机体的重要组成部分,关乎着动物体的生命健康,维持着生命的活性,是新陈代谢的重要组成物。尤其脂肪酸还扮演者信号分子的角色,参与着体内的生命代谢和重要的生物学过程。脂肪的代谢,合成,影响着牛肉的品质,如口感,大理石花纹,牛的机体正常发育,和牛的体质健康,从而也影响着牛的生理健康。肉牛的体尺,体重,胴体重也是影响肉牛产量和品质的一个重要标志。所以这次试验选择了该段与脂肪表达和调控相关的基因现严格选取了日龄相近,生活和词养环境相同的秦川牛公牛为试验材料,提取DNA,利用检测该段基因的多态位点,分析出不同的基因型,研究其显隐性和试验用牛测得性状的一个关联,研究结果为:1、利用PCR-RFLP技术,找出了秦川牛说0叹堪因的三个突变位点,第一个突变位点g.82409AG位于第六条染色体16458977位置;第二个突变位点g.82508OA位于第六条染色体16459076位置,第三个突变位点g.3084AG位于第六条染色体的16379652位置。前两个位于编码区(第二外显子),第三个位于非编码区(第一内含子)。2、三个突变位点均为中度多态性(0.25P0.5)。g.82409AG,g.82508GA处于Hardy-Weinberg极度不平衡状态;g.3084AG处于Hardy-Weinberg平衡状态。3、g.82409AG位点的三个基因型中A A型和B B型对秦川牛的体高影响显著(P=0.043),对体长,腰角宽,体重,胴体重影响不显著(P0.05);g.82508GA对秦川牛的体高,体长,腰角宽的有不同程度的影响,且差异显著(P0.0i),对体重和胴体重影响不显著(p0.05);g.3084AG对秦川牛的体高,体长,腰角宽影响不显著(P0.05),对体重和胴体重影响显著(P0.05)。三个位点产生的不同基因型对牛的肌肉,脂肪组织,或者骨骼的发育有着显著影响,可以试着作为分子辅助选择标记的一个位点,用来研究秦川牛肉品质改善和体型发育。
[Abstract]:VLFAE 6 is the sixth member of the ELOVLs family. There are 5 exons and 4 introns. 10 the function of sigh 6 is mainly manifested in the control and expression regulation of the genes of the C 16 saturated and unsaturated fatty acids family into 18C fatty acids. Fatty acids, in particular, are important components of the body, vital to the life and health of animals, vital to life, and an important component of metabolism. In particular, fatty acids also play the role of signaling molecules. The metabolism and synthesis of fat affect the quality of beef, such as taste, marbling, the normal development of cattle, and the health of cattle. This also affects the physical health of cattle. The body size, body weight of beef cattle, Carcass weight is also an important indicator of beef yield and quality. Qinchuan cattle bull, which has the same living and breeding environment, was used as experimental material to extract DNA. By detecting the polymorphic loci of this gene, different genotypes were analyzed, and an association between the obvious recessive traits and the traits measured by the test cattle was studied. The result of the study was: 1. Using PCR-RFLP technique, we found three mutation sites in Qinchuan cattle. The first mutation locus (g.82409 AG) was located at 16458977 positions on 6th chromosomes, and the second mutation locus (g.82508OA) was located at 16459076 locus of 6th chromosomes. The third mutation locus, g.3084AG, is located at 16379652 of 6th chromosomes. The first two are located in the coding region (exon 2) and the third in the non-coding region (intron 1. 2). The three mutation sites are all moderately polymorphic, 0.25P0.5, g.82409AGG. 82508GA is located at the Hardy-Weinberg pole. In the three genotypes of Hardy-Weinberg equilibrium, A and B have significant effects on body height and body length of Qinchuan cattle. The waist width, body weight and carcass weight had no significant effects on the body height, body length and waist angle width of Qinchuan cattle, and the difference was significant (P 0. 0. 0), but had no significant effect on body weight and carcass weight of Qinchuan cattle. The influence of waist angle width on body weight and carcass weight was not significant. The different genotypes produced at three loci had significant effects on the development of muscle, adipose tissue, or bone of cattle. It can be used as a site of molecular-assisted selection marker to study the improvement of beef quality and the development of body shape in Qinchuan beef.
