PrRP-R在绵羊睾丸和其他组织中的表达与定位分析

【摘要】:为探讨促乳素释放激素受体(prolactin releasing peptide receptor,PrRP-R)基因在不同发育时期绵羊睾丸和不同组织(心脏、肝脏、脾脏、肺脏、肾脏、背最长肌、股二头肌、下丘脑、垂体、松果腺)中表达的差异,以0,2,5,12,24月龄的小尾寒羊为研究对象,应用荧光实时定量PCR(quantitative real-time PCR)和免疫组化(immunohistochemistry)技术分别分析了PrRP-R基因在组织中的差异表达与定位。结果显示:PrRP-R基因在睾丸与心脏、肝脏、脾脏、肺脏、肾脏、背最长肌、股二头肌、下丘脑、垂体、松果腺之间的表达差异极显著(P0.01),股二头肌、垂体、松果腺之间表达差异不显著(P0.05),但背最长肌、下丘脑、垂体之间表达差异显著(P0.05);0,2,5,12,24月龄的睾丸组织中均有一定量的表达,24月龄时表达最高(P0.01);PrRP-R在0~24月龄绵羊睾丸组织中呈阶段性表达,0,2,5月龄时,可观察到PrRP-R在精原细胞中到阳性表达;12,24月龄时,在初级精母细胞和次级精母细胞中检测到强阳性表达。结果表明:PrRP-R在不同的组织和睾丸中表达并存在一定的差异,可能对绵羊的繁殖性能具有重要的调控作用。
[Abstract]:To investigate the expression of prolactin releasing peptide receptor PrRP-RR gene in testis and tissues (heart, liver, spleen, lung, kidney, longissimus dorsi, biceps femoris, hypothalamus, pituitary) of sheep at different developmental stages. The difference of expression in pineal gland was studied in small tail Han sheep aged 24 months. The differential expression and localization of PrRP-R gene in tissues were analyzed by fluorescence real-time quantitative PCR(quantitative real-time and immunohistochemical histochemistry.The results showed that the PrRP-R gene was expressed in testis and heart, liver, spleen, lung, kidney, longissimus dorsi muscle and biceps femoris. The difference of expression among hypothalamus, pituitary and pineal gland was very significant (P 0.01). There was no significant difference in expression between biceps femoris, pituitary and pineal gland, but the longest dorsal muscle and hypothalamus. There was a significant difference in the expression of PrRP-R between the pituitary gland and the testis at 24 months of age. The highest expression of PrRP-R was found in the testis at 24 months of age. The expression of PrRP-R in the testis of 0 to 24 months old sheep showed a periodic expression at the age of 0 to 24 months, and the expression of PrRP-R in the testis of sheep at the age of 0 to 24 months was at the age of 0 to 2 months. Strong positive expression of PrRP-R was observed in primary spermatocytes and secondary spermatocytes at 24 months of age. The results showed that there were some differences in the expression of PrRP-R in different tissues and testis. It may play an important role in regulating the reproductive performance of sheep.
