酵母菌微生态制剂制备工艺研究及应用

【摘要】:本试验选取3株酵母菌(热带假丝酵母R1、酿酒酵母N2和产朊假丝酵母C3),以单因素优化试验为基础,利用固态发酵方式分别对其进行生物活性测定,确定最佳发酵时间、最佳接种量及最佳料水比,筛选出较为经济适用且利用率高的固态培养物,确定其最佳添加比例。通过L9(34)正交试验进行较为系统的优化研究,最终确定3株酵母菌生长的最适发酵条件和最佳培养基,以期获得高产活性酵母菌微生态制剂的制备工艺。同时对研究的产品进行动物生产实践应用,对其饲喂效果进行验证。试验结果如下:(1)在发酵温度为30℃、自然pH条件下,菌株R1最佳发酵时间为48h、接种量为11%、料水比为1:0.75;菌株N2最佳发酵时间为72h、接种量为7%、料水比为1:1.25;菌株C3最佳发酵时间为48h、接种量为11%、料水比为1:1.25。(2)菌株R1、N2、C3固态发酵培养基的最佳碳源种类为甘油,最佳氮源种类为尿素;菌株R1最佳固态发酵培养基配方为麸皮与甘油(g)8.5:1.5、尿素0.5%;菌株N2最佳固态发酵培养基配方为麸皮与甘油(g)9:1、尿素0.5%、磷酸二氢钾0.2%;菌株C3最佳固态发酵培养基配方为麸皮与甘油(g)8.5:1.5、尿素0.5%、磷酸二氢钾0.4%。在优化后的最佳发酵条件及培养基配方下进行固态发酵,菌株R1的活菌数可达32.94× 108个/g,比优化前提高2.26倍;菌株N2的活菌数可达8.60×108个/g,比优化前提高了 6.32倍;菌株C3的活菌数可达2.66× 108个/g,比优化前提高了 1.09倍。(3)日粮中添加复合酵母菌制剂可以有效提高肉羊的整体生产性能,两个时间段内试验肉羊的平均日增重分别为164.89g/d、236.45g/d。与对照组相比,肉羊平均日增重提高了 48.19%,料重比降低了 39.01%。
[Abstract]:Three yeast strains (Candida tropicalis R1, Saccharomyces cerevisiae N2 and Candida prion C3N) were selected in this experiment. Based on the single factor optimization experiment, the bioactivity of the yeast was determined by solid state fermentation, and the optimum fermentation time was determined. The optimum inoculation amount and the best ratio of material to water were used to select the more economical and high utilization ratio of solid state culture, and the optimum proportion of solid culture was determined. The optimization was studied systematically by means of L9 / 34) orthogonal test. Finally, the optimum fermentation conditions and culture medium for the growth of three yeast strains were determined, in order to obtain the preparation process of microecological preparation of yeast with high yield and activity.
