LHR在雌性牦牛生殖器官及肾脏中的表达

【摘要】:为了探讨牦牛肾脏中黄体生成素受体(luteinizing hormone receptor,LHR)的表达情况及其与卵巢中LHR表达的相似性,本研究检测了LHR在牦牛肾脏和卵巢中的表达量。为了探讨LHR基因在牦牛发情周期不同阶段对其生殖器官的调节作用,本研究检测了LHR基因在牦牛发情周期不同阶段生殖系统中的表达差异,并分析了出现差异变化的原因。研究参照黄牛LHR基因序列、牦牛β-actin基因序列和GAPDH基因序列设计特异性引物。选取健康的雌性牦牛为研究对象,利用实时荧光定量PCR(RT-qPCR)法分析牦牛肾脏和卵巢组织中LHR基因的相对表达量,并运用免疫组织化学法对LHR蛋白在牦牛肾脏和卵巢组织中进行表达和定位研究。选取健康的处于卵泡期和黄体期的雌性牦牛为研究对象,利用RT-qPCR法分析牦牛发情周期不同阶段生殖系统中LHR基因的相对表达量。研究结果表明:(1)牦牛肾脏具有表达LHR的特性,其LHR基因的相对表达量是卵巢的79.75倍。(2)LHR在牦牛肾脏组织中的近曲小管和远曲小管呈阳性反应,在卵巢组织中的卵泡颗粒层呈明显的阳性反应,在牦牛肾脏和卵巢中的表达差异极显著(P0.01)。(3)LHR基因在牦牛发情周期不同阶段生殖系统中的表达量存在差异,卵巢和输卵管中卵泡期LHR基因的表达量高于黄体期,子宫中LHR基因的表达量黄体期高于卵泡期。从本研究可以看出,牦牛的肾脏组织与卵巢组织一样,均具有表达LHR的特性,提示牦牛肾脏可通过调节LHR参与其繁殖性能。牦牛在发情周期不同阶段生殖系统中LHR基因的表达量存在差异,提示LHR在牦牛的繁殖过程中对生殖系统具有重要的调节作用。
[Abstract]:To investigate the expression of luteinizing hormone receptor (LHR) in yak kidney and its similarity with LHR expression in ovary. In this study, we examined the expression of LHR in the kidney and ovary of yak. In order to investigate the regulation of LHR gene on the reproductive organs of yak at different stages of estrous cycle, The purpose of this study was to detect the difference of LHR gene expression in the reproductive system of yak at different stages of estrous cycle, and to analyze the causes of the difference. Specific primers were designed for 尾 -actin gene sequence and GAPDH gene sequence of yak. The relative expression of LHR gene in kidney and ovary of yak was analyzed by real-time fluorescence quantitative PCR RT-qPCRmethod. The expression and localization of LHR protein in the kidney and ovary of yak were studied by immunohistochemical method. The healthy female yak in follicular and luteal phase was selected as the study object. RT-qPCR method was used to analyze the relative expression of LHR gene in the reproductive system of yak at different stages of estrous cycle. The results showed that the kidney of yak had the characteristic of expressing LHR. The relative expression of LHR gene was 79.75 times higher than that of ovaries. The proximal tubules and distal convoluted tubules in the kidney tissues of yak showed positive reaction, and the follicular granular layers in the ovarian tissues showed obvious positive reaction. There were significant differences in the expression of P0.01LHR gene in yak kidney and ovaries. The expression of LHR gene in oviducts and oviducts was higher than that in luteal phase in different stages of oestrous cycle of yak, and the expression of P0.01LHR gene in oviduct and oviduct was higher than that in luteal phase. The expression of LHR gene in uterus was higher in luteal phase than in follicular phase. The results suggested that the kidney of yak could participate in the reproductive performance by regulating LHR. There were differences in the expression of LHR gene in the reproductive system of yak at different stages of estrous cycle, suggesting that LHR plays an important role in regulating the reproductive system of yak.
