仔猪均匀度遗传参数估计及仔猪初生重与血清皮质醇、胰岛素含量的相关性研究

【摘要】:研究显示,仔猪均匀度对养猪业有极大影响,血清皮质醇及胰岛素含量可能与仔猪的初生重相关。本研究采用动物模型REML(约束最大似然)方法对大白猪8个繁殖性状的遗传参数进行估计,利用SAS9.1统计软件的GLM程序分析年份、配种季节、胎次对8个繁殖性状的影响,采用Duncan,s法进行各固定效应间繁殖性状的多重比较;采用放射免疫分析法检测不同初生重仔猪血清皮质醇含量及胰岛素含量。旨在为育种工作提供仔猪均匀度遗传参数数据支持;为探索皮质醇及胰岛素对初生重的影响起参考作用。1.仔猪均匀度遗传参数估计利用5种统计模型对大白猪窝内仔猪初生重变异系数(CV0)、窝内仔猪21日龄重变异系数(CV3)、总产仔数(TNB)、产活仔数(NBA)、产死仔数(NBD)、初生窝重(TLW0)、21日龄窝重(TLW3)及妊娠周期(GL)进行遗传参数估算。经比较,应用模型V(包括个体加性遗传效应,母体遗传效应、永久环境效应、与配公猪效应、母猪所在窝效应)得到的残差方差所占比例小。因此本实验采用模型V进行繁殖性状遗传参数估计。遗传参数估计结果表明:TNB,NBA,NBD,CV0,CV3,TLW0,TLW3,GL遗传力分别为0.14,0.13,0.07,0.07,0.02,0.11,0.08,0.09。TNB与NBA、TNB与TLW0、TNB与TLW3、NBA与TLW0、NBA与TLW3、CV0与CV3、TLW0与TLW3之间存在明显的遗传正相关(遗传相关分别为0.93,0.61,0.61,0.65,0.73,0.65,0.91);TNB与CV0、TNB与CV3、CV0与NBD之间遗传相关分别为0.29,0.17,0.22。固定效应方差分析表明:年份对TNB和CV0影响不显著(P0.05),对其余性状的影响都达到极显著(P0.01);配种季节对TNB、NBA、CV3影响不显著(P0.05),对CV0影响达显著水平(P0.05),对TLW0,TLW3和GL影响达到极显著水平(P0.01);胎次对对TNB、NBA、NBD的影响达显著水平(P0.05),对CV0、CV3、TLW0、TLW3及GL影响极显著(P0.01)。产仔数对所有性状影响都达到极显著水平(P0.01)。繁殖性状最小二乘均值表明:随着胎次增加,TNB、NBA、TLW0、TLW3呈现出相同的变化规律,第一胎最低,以后逐渐上升,到3-4胎时候达到最大,然后又开始下降。季节对繁殖性状的影响表现为冬季生产性能最佳;随着产仔数的增加,CV0、CV3、NBD也增加,而MBW0、MBW3、GL却下降;随着产仔数增加,初生重小于1 Kg仔猪所占比例增加,体重大于1.8 Kg仔猪所占比例下降。2.血清皮质醇和胰岛素含量水平与仔猪初生重相关性分析实验选取10窝大白猪,每窝包括2头低初生重(1.0 Kg)2头高初生重(1.2 Kg),共40头初生仔猪作为实验样本。仔猪出生后前腔静脉采血,离心后提取血清,采用放射免疫分析法检测血清中皮质醇含量及胰岛素含量。研究结果表明:高出生重组和低出生重组血清皮质醇量分别为214.72(ng/ml),267.87(ng/ml),两组之间差异极显著(P=0.0083);高出生重组和低出生重组血清胰岛素含量分别为28.67(m IU/L),20.92(m IU/L),两组间差异显著(P=0.0284)。血清皮质醇含量与初生体重之间存在显著负相关(rg=-0.32),血清胰岛素含量与体重之间存在正相关(rg=0.23),但是二者之间相关性不显著(P=0.09)。实验结果为探索皮质醇及胰岛素对初生重的影响提供参考。
[Abstract]:Research shows that piglet uniformity to the pig industry has great influence, and the birth weight of piglets serum cortisol and insulin content. This study used the animal model of REML (restricted maximum likelihood) genetic parameters of 8 reproductive traits in large white pigs estimation method, analysis of the year, the use of SAS9.1 statistical software GLM program of breeding season. Effect of parity on 8 reproductive traits by using Duncan, s multiple comparison method of reproductive traits of fixed effects; radioimmunoassay was used to measure the different birth weight of piglets serum cortisol and insulin content. In order to provide piglets for breeding data uniformity and genetic parameters of support; to explore the effects of insulin and cortisol the birth weight of the piglets reference.1. uniformity of genetic parameter estimation of large white nest piglets weight variation coefficient using 5 statistical model (CV0), wo Zi 21 day old pig weight coefficient of variation (CV3), total litter size (TNB), number born alive (NBA), produce litter (NBD), birth weight (TLW0), litter weight at 21 days (TLW3) and pregnancy period (GL) of genetic parameter estimation. By comparison, application the V model (including individual additive genetic effects, maternal genetic effect and permanent environmental effect, and with the boar sows effect, where litter effect) residual variance obtained small proportion. Therefore this experiment adopts the V model to reproduce the genetic parameters estimation of genetic parameters of traits. The results showed that: TNB, NBA, NBD, CV0. CV3, TLW0, TLW3, GL and 0.14,0.13,0.07,0.07,0.02,0.11,0.08,0.09.TNB respectively, heritability of NBA, TNB and TLW0, TNB and TLW3, NBA and TLW0, NBA and TLW3, CV0 and CV3, there were significant genetic positive correlation between TLW0 and TLW3 (genetic correlation = 0.93,0.61,0.61,0.65,0.73,0.65,0.91); TNB and CV0, TNB and CV3, between CV0 and NBD genetic correlation respectively. Show that the 0.29,0.17,0.22. variance analysis of fixed effects: the effect of TNB and CV0 years (P0.05), no significant effects on other traits were at significant (P0.01); breeding season on TNB, NBA, CV3 had no significant effects on CV0 (P0.05), significant effects (P0.05), TLW0, TLW3 and GL influence reached a significant level (P0.01); parity on to TNB, NBA, NBD significant effects (P0.05), CV0, CV3, TLW0, TLW3 and GL had significant effect (P0.01). Litter size were all highly significant for all traits affecting reproductive traits (P0.01) showed that the least squares mean. With the increase of TNB, NBA, parity, TLW0, TLW3 showed the same variation, the first minimum, after rising to 3-4 when the tire reaches the maximum, and then began to decline. The seasonal effect on reproductive traits for winter production best; with the increase of litter size of CV0, CV3, NBD increased, while MBW0, M BW3, GL decreased; with the increase of litter size, birth weight less than 1 Kg increase in the proportion of piglets, piglets weight greater than 1.8 Kg the proportion of.2. analysis of serum cortisol and insulin levels and birth weight of the piglets correlation experiment selected 10 large white nest down, including 2 low birth weight per litter (1 Kg) 2 heads the birth weight (1.2 Kg), a total of 40 newborn piglets as the experimental samples. The piglets born after blood samples were collected after centrifugation, extract serum, cortisol and insulin content in serum was measured by radioimmunoassay. The analysis results show that the high born restructuring and low birth weight were 214.72 recombinant serum cortisol (ng/ml) (ng/ml, 267.87), significant differences between the two groups (P=0.0083); high birth and low birth of recombinant insulin in recombinant were 28.67 (m IU/L), 20.92 (m, IU/L) between the two groups were significant difference (P=0.0284). The content of serum cortisol There was a significant negative correlation between birth weight and body weight (rg=-0.32). There was a positive correlation between serum insulin and body weight (rg=0.23), but the correlation between the two groups was not significant (P=0.09). The results of the experiment provide a reference for exploring the influence of cortisol and insulin on the birth weight.
