染色质重塑因子BPTF、BRG1在不同毛色绵羊皮肤的表达与定位

【摘要】:旨在研究染色质重塑因子BPTF、BRG1在不同毛色绵羊皮肤的表达与定位。采集黑色和白色各3只绵羊的背部皮肤组织,用qRT-PCR检测不同毛色皮肤中BPTF和BRG1mRNA相对表达量,免疫组化技术定位分析,Western blotting半定量研究BPTF和BRG1蛋白相对表达量。qRT-PCR结果表明,BPTF基因在黑色绵羊皮肤中的mRNA相对表达量显著高于白色绵羊(P0.05),BRG1基因在黑色绵羊皮肤中的mRNA相对表达量极显著高于白色绵羊(P0.01);Western blotting结果表明,黑色绵羊的皮肤中BPTF蛋白表达量显著高于白色绵羊(P0.05),BRG1蛋白表达量极显著高于白色绵羊(P0.01);免疫组化结果表明,BPTF蛋白和BRG1蛋白在绵羊皮肤中毛囊的毛根鞘及毛球部均有表达。综上表明,BPTF和BRG1基因可能参与绵羊毛色形成过程。
[Abstract]:The aim of this study was to investigate the expression and localization of chromatin remodeling factor BPTF- BRG1 in sheep skin.The back skin tissues of 3 black sheep and 3 white sheep were collected, and the relative expression of BPTF and BRG1mRNA were detected by qRT-PCR.Expression of BPTF and BRG1 protein in Black Sheep skin by Immunohistochemistry and Semi-quantitative Analysis of Western blotting. QRT-PCR results showed that the relative expression of mRNA gene in black sheep skin was significantly higher than that in black sheep skin.The relative expression of mRNA was significantly higher than that of P0.01 in white sheep.The expression of BPTF protein in the skin of black sheep was significantly higher than that in the white sheep, and the expression of BPTF protein and BRG1 protein in the hair follicle of sheep was significantly higher than that in the white sheep P0.01.The immunohistochemical results showed that the expression of BPTF protein and BRG1 protein in the hair follicle and hair follicle of the sheep were all positive.It is concluded that BPTF and BRG1 genes may be involved in the process of wool color formation.
