大通犊牦牛组织中线粒体SDH活性与其亚基mRNA表达的测定

【摘要】:为探讨高原低氧环境下大通犊牦牛线粒体琥珀酸脱氢酶(SDH)在不同组织中的表达模式以及SDH活性与其亚基mRNA表达量之间的关系,采用酶联免疫分析法(ELISA)对犊牦牛心肌、骨骼肌、大脑、肝脏组织中线粒体SDH活性进行测定;实时荧光定量PCR(Real-Time PCR)检测组织中Sdha、Sdhb、Sdhc和Sdhd亚基mRNA表达量。结果表明:大通犊牦牛大脑、心肌、骨骼肌组织中线粒体SDH活性显著高于乐都犊牦牛;大脑组织中线粒体Sdh4个亚基mRNA表达量极显著高于乐都犊牦牛。结论:含野血大通犊牦牛组织线粒体SDH有氧代谢功能强于乐都犊牦牛;大脑组织线粒体Sdh4个亚基转录水平较高;大通犊牦牛大脑组织线粒体SDH活性与Sdh4个亚基mRNA表达量呈正相关。
[Abstract]:In order to investigate the expression pattern of mitochondrial succinate dehydrogenase (SDH) in different tissues and the relationship between the activity of SDH and the expression of its subunit mRNA in Datong calf yak under high altitude hypoxia, the myocardial and skeletal muscles of calf yak were treated by Elisa.The activity of mitochondrial SDH was measured in brain and liver, and the expression of Sdhagsdhbhc and Sdhd subunit mRNA was detected by real-time fluorescence quantitative PCR(Real-Time.The results showed that the activity of mitochondrial SDH in brain, myocardium and skeletal muscle of Datong calf yak was significantly higher than that of Ledong calf yak, and the mRNA expression of mitochondrial Sdh4 subunit in brain tissue was significantly higher than that in Ledong calf yak.Conclusion: the aerobic metabolism function of mitochondrial SDH in yak with wild blood is stronger than that in Ledong calf, and the transcription level of mitochondrial Sdh4 subunit in brain tissue is higher than that in Ledong calf yak.There was a positive correlation between mitochondrial SDH activity and mRNA expression of Sdh4 subunits in the brain of Datong calf yak.
