奶牛黄体囊肿的蛋白质组学研究

【摘要】:为了研究奶牛黄体囊肿的发病机理及发病时的生物学标记物,利用蛋白质组学经典的双向电泳(2-DE)技术,检测和分析奶牛黄体囊肿组织和正常黄体组织间的差异蛋白,通过考马斯亮蓝染色后用PDQuest8.0软件检测并分析差异蛋白点。经MALDI TOF/TOF-MS/MS串联质谱鉴定,共发现6种差异表达的蛋白,分别为:α-1-酸性糖蛋白(AGP),胶质纤维酸性蛋白(GFAP),核磷蛋白(NPM),泛素羧基末端水解酶(UCH),牛血清白蛋白晶体结构A链(Chain A),丝氨酸蛋白酶抑制蛋白A3-5(serpin A3-5)。其中,AGP、GFAP、serpin A3-5、NPM和UCH在囊肿组织中高表达,Chain A只在正常组织中表达。通过分析这些蛋白在组织间的表达差异,发现其可能与黄体囊肿的发生和囊肿发生后激素分泌异常有关。黄体囊肿差异蛋白的发现为奶牛黄体囊肿临床诊断及发生机理的研究提供了科学依据。
[Abstract]:In order to study the pathogenesis and biological markers of luteal cyst in dairy cattle, differential proteins were detected and analyzed between the tissues of luteal cyst and normal corpus luteum by using the proteomics technique of two-dimensional electrophoresis (2-DEE).The differential protein spots were detected and analyzed by PDQuest8.0 software after Coomassie brilliant blue staining.Six differentially expressed proteins were identified by MALDI TOF/TOF-MS/MS tandem mass spectrometry.They were 伪 -1-acidic glycoprotein (AGP), glial fibrillary acidic protein (GFAP), nucleophosphorus protein (NPP), ubiquitin carboxyl terminal hydrolase (UCHN), crystal structure of bovine serum albumin (BSA) A chain chain An, serine protease inhibitor protein (A3-5(serpin A3-5).UCH and UCH were highly expressed in cysts only in normal tissues.By analyzing the differences in the expression of these proteins among tissues, it was found that these proteins might be related to the occurrence of luteal cyst and the abnormal hormone secretion after cysts.The discovery of differential protein in luteal cyst provides scientific basis for clinical diagnosis and mechanism of luteal cyst in dairy cattle.
