丝兰提取物对肉仔鸡抗氧化和免疫功能及其相关基因表达的影响

【摘要】:本论文通过在日粮中添加不同浓度的丝兰提取物(Yucca schidigera extract,YSE),研究YSE对肉仔鸡抗氧化和免疫功能的影响,探讨YSE在肉仔鸡日粮中的应用效果,为确定肉仔鸡日粮中YSE的适宜添加量提供理论依据。试验选取体重相近的健康15日龄的AA肉仔鸡128只,随机分为4个处理组,每个处理4个重复,每个重复8只鸡(公母各半)。将玉米豆粕型日粮作为基础日粮,对照组饲喂基础日粮,3个YSE添加组分别添加100、200、300 mg/kg的YSE。试验期28 d,分为试验前期(15~28 d)和试验后期(29~42 d),各14 d。论文共分三个试验内容:试验一:通过测定血清、小肠、肝脏、胸肌样品中抗氧化酶活力及其产物浓度,并测定组织中抗氧化酶基因的相对表达量,研究YSE对于肉仔鸡抗氧化功能的影响。.结果表明:在14 d,血清中总抗氧化能力(T-AOC)和过氧化氢酶(CAT)水平均在200 mg/kg YSE添加剂量时提高(P=0.031,P=0.009),血清中超氧化物歧化酶(SOD)活性在所有YSE添加水平均升高(P=0.014);200mg/kg的YSE添加水平提高了肝脏中CAT活力及其基因表达(P=0.035,P=0.003);十二指肠和回肠中T-AOC(P=0.039,P=0.031)、谷胱甘肽过氧化物酶(GPx)(P=0.016,P=0.045)和SOD(P=0.001,P=0.023)水平在100mg/kgYSE添加水平时均升高;100mg/kgYSE上调了回肠中SOD和GPx的基因表达(P=0.018,P=0.003),回肠中CAT的活力及其基因相对表达量在100 mg/kg添加水平时升高(P0.001,P=0.028)。在28 d,血清中T-AOC水平在200和300 mg/kg剂量时上升(P=0.006),但MDA含量仅在300 mg/kg时显著降低(P=0.019);肝脏T-AOC水平在300 mg/kg YSE添加剂量时显著提高(P=0.001);300mg/kgYSE添加量显著升高了肝脏中GPx和SOD活性(P=0.002,P=0.049),并上调了其GPx的基因表达(P=0.028);肝脏MDA浓度在300 mg/kg剂量组显著降低(P=0.006);十二指肠SOD活力在100mg/kg时上升(P=0.039);所有YSE添加组均能提升回肠中GPx水平(P=0.022)。300mg/kgYSE添加能够显著提高胸肌T-AOC的水平(P=0.017)。试验二:通过测定血清中免疫细胞因子浓度及新城疫抗体滴度,探讨在肉仔鸡日粮中添加YSE对其免疫功能的影响。结果表明:在14 d,血清中白介素(IL)-6浓度在 200 和 300 mg/kg YSE 添加水平时降低(P0.001);在 28 d,100 mg/kg YSE浓度降低了 IL-4浓度(P=0.002),提高了 IL-6和γ干扰素(IFN-γ)的浓度(P=0.004,P=0.026);在14 d和28 d,新城疫抗体滴度在100 mg/kg YSE添加水平时升高(P0.001,P=0.032)。试验三:通过测定血清中一氧化氮(NO)及诱导型一氧化氮合酶(iNOS)水平,及肝脏、脾脏、小肠中iNOS的基因表达,从NO途径探究YSE对于肉仔鸡免疫及抗氧化功能的影响。结果表明:在14d,100mg/kgYSE促进了血清中NO含量和iNOS的活性(P=0.032,P=0.048),200 mg/kg YSE上调了回肠中iNOS的基因表达(P=0.031),肝脏和脾脏中iNOS基因表达在100 mg/kg YSE添加水平呈现上调趋势(P=0.081,P=0.075);在28d,与对照组相比,YSE处理组血清中NO和iNOS浓度,组织中iNOS的基因表达均未出现显著差异。本试验结果表明,在日粮中添加YSE能够提升肉仔鸡的抗氧化及免疫功能。100、200和300 mg/kg YSE添加水平能够通过提高不同组织的抗氧化指标来提升肉仔鸡机体的抗氧化功能;100mg/kg的YSE添加能够促进肉仔鸡的免疫功能;100mg/kg的YSE添加能够通过提升血清中NO浓度及iNOS活性从而提升肉仔鸡的抗氧化和免疫功能。
[Abstract]:In this paper, by adding different concentrations of yucca extract in diets (Yucca schidigera, extract, YSE), the effects of YSE on antioxidant and immune function of broilers, to explore the application effect of YSE in broiler diets, as determined in broiler diets the appropriate addition amount of YSE and provide a theoretical basis for the similar weight. 15 day old 128 AA broiler chickens were selected, randomly divided into 4 groups with 4 replicates of 8 chicks per replicate (male and female). The corn soybean meal diet as basal diet. The control group was fed basal diet, 3 YSE supplementation groups were added 100200300 mg/kg YSE. test of 28 d, divided into pre test (15~28 d) and the end of the experiment (29~42 d), each 14 d. of the experiment was divided into three parts: test one: by determination of serum, small intestine, liver, antioxidant enzyme activity and product concentration in sample and determination of muscle, tissue The relative expression of anti oxidase gene, effect of YSE on the antioxidant function in broilers. The results showed that at 14 d, the total antioxidant capacity in serum (T-AOC) and catalase (CAT) levels were increased in 200 mg/kg YSE added dose (P=0.031, P=0.009), serum superoxide dismutase (SOD) activity increased in all YSE levels (P=0.014); 200mg/kg YSE added to improve the level of CAT activity and gene expression in the liver (P=0.035, P=0.003); T-AOC in the duodenum and ileum (P=0.039, P=0.031), glutathione peroxidase (GPx) and Gu Guanggan (P=0.016, P=0.045) and SOD (P=0.001, P=0.023) level increased in 100mg/kgYSE when the addition level of 100mg/kgYSE increased; the expression of SOD and GPx in the ileum of genes (P=0.018, P=0.003), relative activity and mRNA expression in the ileum of CAT increased at 100 mg/kg when the addition level of (P0.001, P=0.028). In 28 d, blood In the T-AOC level increased in 200 and 300 of the dose of mg/kg (P=0.006), but the content of MDA in only 300 mg/kg significantly decreased (P=0.019); liver T-AOC levels increased significantly in the 300 mg/kg YSE adding dose (P=0.001); the amount of 300mg/kgYSE significantly increased GPx and SOD activity in liver (P=0.002, P=0.049). Two. Test: through the determination of serum cytokine concentrations and Newcastle disease antibody titer, investigated in broiler diets the effect of YSE on immune function. The results showed that at 14 d, serum interleukin (IL) -6 concentration at 200 and 300 mg/k G YSE decreased when the addition level (P0.001); in 28 d, 100 mg/kg YSE concentration decreased the concentration of IL-4 (P=0.002), increased IL-6 and interferon gamma (IFN- gamma) concentration (P=0.004, P=0.026); in 14 d and 28 d, Newcastle disease antibody titer increased at 100 mg/kg when the addition level of YSE (P0.001, P=0.032). Experiment three: the determination of serum nitric oxide (NO) and inducible nitric oxide synthase (iNOS) levels, and the liver, spleen, intestinal iNOS gene expression, to explore the impact of YSE on immune function and antioxidation of broiler chickens from the NO pathway. The results showed that: in 14d, 100mg/kgYSE promoted NO content and iNOS activity in serum (P=0.032, P=0.048), 200 mg/kg YSE up-regulated the expression of iNOS gene in the ileum (P=0.031, iNOS) in the liver and spleen gene expression level increased in adding 100 mg/kg YSE (P=0.081, P= 0.075); in 28d, compared with the control group, YSE treatment group in serum N O and iNOS concentration, the expression of iNOS gene had no significant difference. The test results show that the addition of YSE can enhance the antioxidant and immune function of.100200 broilers and 300 mg/kg YSE levels can increase the antioxidant index of different tissues to enhance the body's antioxidant function in Broilers Diets; 100mg/kg YSE add can promote the immune function of broilers; 100mg/kg YSE is added to be able to enhance the concentrations of serum NO and iNOS activity so as to enhance the antioxidant and immune function of broilers.
