妊娠后期饲粮添加共轭亚油酸影响母猪初乳IgG的作用机理

【摘要】:论文通过两个饲养试验和两个细胞试验研究妊娠后期饲粮添加共轭亚油酸(Conjugated linoleicacid,CLA)对初产和经产母猪繁殖性能、初乳产量和免疫球蛋白G(ImmunoglobulinG,IgG)含量的影响及其可能的分子作用机制。试验一,选用48头平均体重为257.6± 1.5 kg,平均胎次为5.5±0.1胎的经产长白×大白杂交母猪,按体重和胎次相近的原则随机分配到4个处理,每个处理12个重复,每个重复1头母猪,从妊娠第85 d到分娩前分别饲喂含0%(对照组)、0.75%、1.50%和2.25%CLA的试验饲粮。结果表明,饲粮中添加共轭亚油酸对经产母猪的窝产仔数、初乳产量及其仔猪的生长性能均无显著影响。与对照组相比,添加共轭亚油酸提高妊娠第112d母猪血清(P0.05)、初乳(P0.01)以及新生仔猪血清(P0.01)中IgG含量,但添加不同CLA 水平的处理间没有显著差异。试验二,选用24头平均体重为170.7 ± 2.3 kg的初产长白×大白杂交母猪,按体重相近的原则分配到4个处理,每个处理6个重复,每个重复1头母猪,从妊娠第85d到分娩前分别饲喂含0%(对照组)、0.75%、1.50%和2.25%CLA的试验饲粮。结果表明,与对照组相比,添加1.5%和2.25%CLA提高初产母猪初乳IgG的含量(P0.05),而添加2.25%CLA可以提高新生仔猪血清中IgG含量(P0.05),降低哺乳仔猪的死亡率(P0.05)。试验三,选用不同浓度(0、12.5、25、50、100和200μM)的顺9,反11(c9,t11)、反10,顺12(t10,c12)共轭亚油酸以及两者的等比例混合物对美洲商陆丝裂原刺激下猪外周血单核细胞(Peripheral blood mononuclear cells,PBMCs)的体外增殖率和IgG生成的影响。结果表明,t10,c12-CLA浓度为12.5和25 μM时促进PBMCs的增殖(P0.05)。当c9,t11-CLA浓度为50μM以上,以及t10,c12-CLA和两种CLA异构体混合物的浓度为100 μM以上时,PBMCs的增殖受到抑制(P0.05)。另外,c9,t11-CLA、t10,c12-CLA及其混合物浓度分别为12.5、12.5和25、25 μM时促进猪外周血单核细胞IgG的生成(P0.05)。试验四,在试验三的基础上进一步研究t10,c12-CLA 对猪 PBMCs 蛋白激酶 C(Protein kinase C,PKC)和细胞因子生成,以及 t10,c12-CLA 和 GW9508 对 IgG 生成和 G 蛋白偶联受体 120(G-protein coupled receptor 120,GPR120)蛋白表达的影响。结果表明,t10,c12-CLA促进猪PBMCs白介素-2和PKC的生成(P0.05),抑制干扰素-γ的生成(P0.05)。t10,c12-CLA及其与GW9508的组合显著提高猪外周血单核细胞IgG的生成(P0.05)。t10,c12-CLA、GW9508以及两者的组合提高猪PBMCsGPR120蛋白的表达(P0.05)。结论,妊娠后期饲粮添加共轭亚油酸可以提高初产母猪和经产母猪初乳IgG的含量,与c9,t11-CLA相比,t10,c12-CLA在促进IgG生成方面可能发挥更重要的作用。
[Abstract]:In this paper, the effects of conjugated linoleic acid on reproductive performance, colostrum yield and immunoglobulin G G (IgG) content in newborn and adult sows were studied by two feeding experiments and two cell tests. In experiment 1, 48 Landrace 脳 big white crossbred sows with average body weight of 257.6 卤1.5 kg and average birth order of 5.5 卤0.1 were randomly assigned to 4 treatments according to the principle of similar body weight and birth order, 12 replicates per treatment and 1 sow per replica. The experimental diet containing 0.75% and 1.50% 2.25%CLA was fed from the 85th day of pregnancy to the beginning of labor. The results showed that the addition of conjugated linoleic acid in diet had no significant effect on litter size, colostrum yield and growth performance of piglets. Compared with the control group, conjugated linoleic acid (CLA) increased the content of IgG in serum of sows (P0.05), colostrums (P0.01) and newborn piglets (P0.01) on the 12th day of gestation, but there was no significant difference between the treatments with different levels of CLA. In experiment 2, 24 Landrace 脳 big white crossbred sows with an average body weight of 170.7 卤2.3kg were selected and assigned to four treatments according to the principle of similar body weight, six replicates per treatment and one sows per replica. The experimental diet containing 0.75% and 1.50% 2.25%CLA was fed from day 85 to delivery. The results showed that addition of 1.5% and 2.25%CLA could increase the content of IgG in colostrum and increase the content of IgG in the serum of newborn piglets, and decrease the mortality of lactating piglets. In experiment 3, the effects of cis-9, anti-11c9t11, anti-10, cis-12t10c12) conjugated linoleic acid and their mixtures on proliferation rate and IgG production of peripheral blood mononuclear cells of porcine peripheral blood monocytes stimulated by American pokeweed mitogen were studied. The results showed that the concentration of C12-CLA was 12.5 渭 M and 25 渭 M, which promoted the proliferation of PBMCs. When the concentration of c9t11-CLA was more than 50 渭 M and the concentration of t10c12-CLA and two CLA isomers was more than 100 渭 M, the proliferation of CLA was inhibited. In addition, the concentration of C9T11-CLAT10C12-CLA and its mixture were 12.5 and 2525 渭 M, respectively, to promote the production of IgG in porcine peripheral blood monocytes (P0.05). In experiment 4, on the basis of experiment 3, we further studied the effects of t10c12-CLA on the production of porcine PBMCs protein kinase (C(Protein kinase) and cytokines, and on the expression of IgG and G protein-coupled receptor (120(G-protein coupled receptor 120-GPR120) by t10c12-CLA and GW9508. The results showed that t10c12-CLA promoted the production of PBMCs interleukin-2 and PKC, inhibited the production of IFN- 纬 and its combination with GW9508 significantly increased the production of IgG in porcine peripheral blood mononuclear cells (P0.05N. T10C12-CLAGW9508) and the combination of the two increased the expression of porcine PBMCsGPR120 protein. Conclusion: supplementation of conjugated linoleic acid in diet in late pregnancy can increase the content of IgG in colostrum of first born sows and sows. Compared with c9t11-CLA, t10c12-CLA may play a more important role in promoting the production of IgG.
