妊娠后期宫内生长限制对蒙古绵羊胎儿肝脏细胞增殖及其相关基因表达的影响

【摘要】:本论文系统的研究了妊娠后期宫内生长限制对蒙古绵羊胎儿肝脏细胞增殖及相关基因表达的影响。选取健康蒙古绵羊(经同期发情受孕)24只。妊娠90 d时,选择与总体平均体重接近的6只母羊进行屠宰,剩余绵羊按体重随机分配到3个营养水平处理组:0.18MJME·BW-0.75·d-1 (RG1:n=6)、0.33 MJME·BW-0.75·d-1 (RG2:n=6)和自由采食(CG:n=6),按以上营养水平饲养至妊娠140 d,将每组6只母羊进行屠宰实验。试验结果表明:(1)妊娠后期限饲母羊导致限饲组胎儿RG2 (p0.05),RG1组(p0.01)胎儿肝脏重,RG1组胎儿肝脏中ⅠCol (p<0.01)、 ⅢCol (p0.05)、ⅣCol(p<0.01) LN (p0.01), HA (p0.05)以及FN(p0.01),RG2组胎儿肝脏FN(p0.01)均显著低于CG组。RG2、RG1组胎儿肝脏组织结构受到严重影响,网状纤维和胶原纤维明显增多变粗。(2) RG2、RG1组胎儿肝脏DNA浓度值、蛋白质/DNA均显著低于CG组(p0.01); RG1组G0/G1期细胞百分比显著高于RG2组(p0.05)、CG组(p0.01);RG2组(p0.05)、RG1组(p0.01)S期细胞百分比均显著低于CG组:且RG2组(p0.05)、RG1组(p0.01)增殖指数均显著低于CG组。(3)RG1组、RG2组CyclinA蛋白的表达量显著低于CG组(p0.05),且RG1组、RG2组CyclinB,,CyclinE蛋白的表达量也显著低于CG组(p0.01),RGl组CyclinD蛋白的表达量显著高于RG2组(p<0.05); RG1、RG2组CDK1蛋白表达量(p<0.05)、CDK2蛋白表达量(p<0.01)、 CDK4蛋白表达量(p0.01)显著低于CG组;RG1组E2F2蛋白表达量显著低于RG2组、CG组(p0.01);RG1组E2F5蛋白表达量显著高于RG2组和CG组(p<0.01); CG、RG2、RGl组胎儿肝脏中CDK1、 CyclinD1、E2F5基因表达量逐渐上调,而CyclinB1、CDK4、E2F1基因表达量逐渐下调。
[Abstract]:The effects of intrauterine growth restriction on the proliferation of fetal liver cells and the expression of related genes in Mongolian sheep were systematically studied in this paper. A total of 24 healthy Mongolian sheep were selected. At the 90th day of gestation, 6 ewes, which were close to the total average body weight, were slaughtered. The remaining sheep were randomly divided into 3 groups according to their body weight: 0. 18MJME BW-0.75 d-1 RG1: nongfong 0.33 MJME BW-0.75 d-1 RG2: nongf6) and free feed CG2: nongf6).
