山羊STAT5A和STAT5B基因表达及遗传变异与其生长性状关联研究

【摘要】:本研究以贵州省地方优良山羊品种——贵州白山羊、贵州黑山羊和黔北麻羊3种山羊作为研究对象,试验山羊共计337只,利用DNA混合池、PCR-SSCP、DNA直接测序等方法研究STAT5A和STAT5B基因单核苷酸多态性,并分别研究了不同山羊品种群体遗传结构、遗传多样性与生长性状的关联分析,以期得到普遍适用于能显著提高山羊各品种个体生长性状的分子遗传辅助标记。同时,本研究还利用q PCR技术分析了贵州白山羊STAT5A和STAT5B基因在9个不同组织中的表达。研究结果如下:1在三种山羊STAT5A和STAT5B基因中分别筛查到3个SNPs2 STAT5A基因C11447T位点对贵州白山羊体高有显著影响,G14264A位点对贵州黑山羊体重、体高和胸围有显著影响,G14320A位点对贵州白山羊体斜长、管围和胸围及贵州黑山羊体重和胸围有显著影响(P0.05)3 STAT5B基因T16159C位点对贵州白山羊体重、体斜长、管围和胸围及黔北麻羊体重、体高、管围和胸围有显著影响,C20844T位点对贵州白山羊体重、体斜长、管围和胸围及贵州黑山羊胸围有显著影响,C21455T位点对贵州白山羊5个生长性状,贵州黑山羊体重和胸围及黔北麻羊体重、体高和体斜长有显著影响(P0.05)4 STAT5A和STAT5B基因在贵州白山羊不同年龄阶段表达量(1)STAT5A周岁羊:肝垂体心下丘脑肺脾小脑大脑肾;(2)STAT5A成年羊:心肺大脑垂体下丘脑小脑脾肝肾;(3)STAT5B周岁羊:肝垂体心脾小脑肺大脑肾下丘脑;(4)STAT5B成年羊:心肺大脑肝垂体肾脾小脑下丘脑。5 STAT5A和STAT5B基因在贵州白山羊周岁羊肝脏和垂体中表达最高,在贵州白山羊成年羊心脏、肺和大脑表达最高,暗示这两个基因是调控山羊早期生长的重要基因,且随着山羊个体的发育成熟,其主要作用和作用部位发生改变,同时这两个基因具有高度的协调性与一致性。
[Abstract]:In this study, three kinds of goats, Guizhou white goat, Guizhou black goat and Qianbei Ma goat, were used as the research objects. A total of 337 goats were tested. Single nucleotide polymorphisms (SNP) of STAT5A and STAT5B genes were studied by DNA mixed pool PCR-SSCP DNA sequencing, and the genetic structure, genetic diversity and growth traits of different goat breeds were analyzed. In order to obtain the molecular genetic assistant markers which can improve the individual growth traits of goat breeds. At the same time, Q PCR technique was used to analyze the expression of STAT5A and STAT5B genes in 9 different tissues of Guizhou White Goat. The results were as follows: 1. Three SNPs2 STAT5A C11447T loci were screened in three goat STAT5A and STAT5B genes, respectively, which had a significant effect on the body height of Guizhou White Goat and the weight of Guizhou Black Goat was affected by G14264A locus. Body height and chest circumference had significant effects on body weight, body length, tube circumference and chest circumference of Guizhou white goats and body weight and chest circumference of Guizhou black goats. The T16159C locus of P0.05 STAT5B gene had significant effects on body weight, body length, tube circumference and chest circumference of Guizhou white goats and body weight of Qianbei Ma sheep. Body height, tube circumference and chest circumference had significant effects on body weight, body oblique length, tube circumference and chest circumference of Guizhou black goat, and C21455T locus on five growth traits of Guizhou white goat. Weight and chest circumference of Guizhou Black Goat and weight of Guizhou Black Goat, Body height and body oblique length have significant effects on the expression of P0.05 STAT5A and STAT5B genes in Guizhou White Goat at different age stages: liver, pituitary, heart, hypothalamus, lung, spleen, cerebellum, kidney, adult sheep: cardiopulmonary pituitary, hypothalamus, cerebellum, spleen, liver, kidney, liver and kidney. ST5B: liver, pituitary, heart, spleen, cerebellar, cerebellar, cerebellar and renal hypothalamus, adult sheep: cardiopulmonary cerebrum, liver, pituitary, kidney, cerebellar hypothalamus, liver, lung, cerebrum, pituitary, kidney and cerebellar hypothalamus showed the highest expression in Guizhou white goat, liver and pituitary. The expression of these two genes in the heart, lungs and cerebrum of Guizhou white goat is the highest, suggesting that these two genes are important genes to regulate the early growth of goats, and their main roles and action sites change with the development and maturation of goat individuals. At the same time, the two genes have a high degree of coordination and consistency.
